宁肠汤对肠易激综合征大鼠结肠黏膜炎性因子IL-10的影响

摘　　要：目的:观察宁肠汤对束缚应激法诱导腹泻型肠易激综合征(IBS)模型大鼠小肠运动功能及大鼠结肠黏膜炎性细胞因子IL-10的影响,探讨宁肠汤在IBS发病机制中可能的作用及临床意义。方法:将60只雄性大鼠随机分为正常对照组、模型对照组、得舒特组、宁肠汤低、中、高剂量组,采用williams改良方法建立束缚应激+适度夹尾刺激致大鼠腹泻模型,观察宁肠汤对大鼠小肠运动功能的影响;通过免疫组化法,对各组大鼠横结肠黏膜进行IL-10表达定量。结果:模型对照组大鼠小肠墨汁推进率较正常对照组明显提高(P<0.05),而各治疗组较模型对照组明显下降(P<0.05);正常对照组IL-10表达量最低,模型对照组大鼠横结肠黏膜IL-10表达量最高,高浓度宁肠汤剂量组相对低浓度宁肠汤剂量组IL-10表达量较低(P<0.05)。结论:宁肠汤对束缚应激大鼠亢进的小肠蠕动有抑制作用,高浓度宁肠汤剂量影响IL-10表达量的作用要优于低浓度宁肠汤组(P<0.05),IL-10的表达量或可以作为衡量IBS药物治疗有效性的生物学标志。Objective： Studying the effect of Ningchang decoction on small intestine＇ s movements and the expres- sion of IL-10 on the colon mucosa of irritable bowel syndrome（IBS） rats resulted from stimulation of proper fettering to explore the potential mechanism and clinical significance. Methods： 60 male SD rats were randomly divided into normal control group, model control group, Dicetel group, low dosage of Ningchang decoction group, medium dosage of Ningchang decoction group and high dosage of Ningchang decoction group. The IBS mice model was induced by proper fettering and tail-clamping on the basis of modified Williams method to observe the effect of Ningchang de- coction on intestinal motility function in rats. And the IL-10 expression of colonic mucosa in each group were evalu- ated by immunobistochemical method. Results： The rate of small intestines＇ ink-proceding of model control group is much faster than that of normal control group（P〈0.05）, while all the treating groups＇ ink-proceding rate are lower than model control group（P〈0.05）. The expression of IL-10 was lowest in normal control group while highest in model con- trol group. And that in high dosage of Ningchang decoction group was lower than low dosage of Ningchang decoction group（P〈0.05）. Conclusion： Ningchang decoction can inhibit the hyper-function of intestinal motility of the diarrhea rats induced by proper fettering. The influence of IL-10 expression of the high dosage of Ningchang decoction group was more significant than that of the low dosage of Ningchang decoction group. The expression of IL-10 can be used as a biological marker to screen the therapeutic effect of IBS drugs.
