摘  要:目的应用同位素标记相对和绝对定量蛋白质组学技iTRAQ数据对获得的蛋白组,采用Uniprot数据库进行基因功能聚类(GO)分析、蛋白分类及路径(Pathway)分析。方法对易栓症患者血清与正常血清标本,进行酶解、标记等步骤处理样本。应用同位素标记相对和绝对蛋白定量法和功能聚类分析法筛查和分析易栓症患者血清和正常血清的差异表达蛋白。结果共有50个差异表达蛋白质被确定,其中包括29个蛋白表达上调和21个蛋白下调。从而总结易栓症危险因素为进一步的研究提供证据。结论 iTRAQ技术和生物信息学方法为筛选出有意义的易栓症标记物提供了可能。Objective:Dentification and comparative quantification between protein samples were performed using isobaric tags for relative and absolute qutification(iTRAQ),which used to analyze gene function cluster(GO),protein classification and path analysis(Pathway). Methods:The serum sample of thrombophilia patients and normal serum samples were treated by enzyme digestion and labeling.To investigate the differentially expressed proteins in serum of patients in Thrombophilia and normal serum using isobaric tagging for relative and absolute protein quantification and gene ontology analysis. Results:Totally,50 differentially expressed proteins were identified in the test,including 29 up-regulated proteins and 21 down-regulated proteins,and sums up the development of thrombophilia risk factors,so as to provide the basis for its further research. Conclusion:The i TRAQ technology and bioinformatics methods provide the possibility to screen out the significant markers of the Thrombophilia.
