摘 要:目的:通过夹脊电针对ALS-SOD1^(G93A)转基因小鼠的干预,观察其对小鼠腰髓脊髓前角运动神经元P38、P-P38表达,旨在证实针刺对神经元细胞的保护作用,为夹脊电针治疗肌萎缩侧索硬化(ALS)提供实验依据。方法:选取ALS-SOD1^(G93A)转基因小鼠18只,按随机数字表法随机将其分为电针组、手针组、模型组,每组各6只,阴性对照组选取同窝野生型小鼠6只。在小鼠出生后第60天开始进行干预。电针组:双侧L_(1~2)、L_(5~6)夹脊穴针刺,疏波刺激;手针组:双侧L_(1~2)、L_(5~6)夹脊穴针刺,每10 min捻转1次;模型组、阴性对照组:同一时间,同一时长抓握、捆绑处理。共4周。小鼠出生120天时进行PP38MAPK、P38MAPK免疫组织化学法检测,并用图像分析仪对每张图片阳性表达的平均光密度(mean density)及累积光密度(IOD)进行测定和统计分析。结果:模型组P-P38、P38表达及P-P38/P38比值均明显比阴性对照组升高,且具有极显著性差异(P<0.01)。与模型组和手针组相比,电针组表达明显降低(P<0.01或P<0.05);与模型组比较,虽然手针组比值有所降低,但无显著性差异(P>0.05)。说明夹脊电针可以明显降低ALS-G93A小鼠腰髓前角运动神经元P-P38、P38表达及P-P38/P38比值。结论:夹脊电针对发病中期的ALS-SOD1^(G93A)转基因小鼠腰髓前角运动神经元P-P38表达有下调作用,抑制P38MAPK的活化,从而起到有效保护神经元细胞的作用。Objective:To observe the expression of P38,P-P38 in motor neuron of anterior horn of lumber spinal cord by electro-acupuncture on Jiaji points in ALS-SOD1^G93A transgenic mice,to prove acupuncture protective effect on neuron and to provide experimental basis for the acupuncture treatment for ALS.Methods:18 ALS-SOD1^G93A transgenic mice were randomly divided into the electro-acupuncture group,the acupuncture group,the model group,with six mice in each group;six wild-type mice were taken as negative control.The mice were intervened starting from 60 days after birth.The electro-acupuncture group was intervened with rarefaction wave on Jiaji points of L(1 - 2),and L(5 - 6);the acupuncture group was intervened with hand acupuncture on Jiaji points of L(1 - 2),and L(5 - 6),the frequency for the needle twirling was once every ten minutes;the model group and the negative control group were intervened with grasping and seizing at the same time for the same time length.The treatment course was four weeks.120 days after birth,the mice were given immunohistochemical test,and the mean density and IOD were analyzed by image analyzer.Results:The expressions of P-P38 and P38,and the ratio of P-P38/P38 in the model group were significantly increased compared to the negative control group(P〈0.01).The expressions was decreased in the electro-acupuncture group compared to those of the model group and the acupuncture group(P〈0.01 or P〈0.05),while there were no significant differences between the acupuncture group and the model group(P〈0.05).It proved that electro-acupuncture could reduce the expressions of P-P38 and P38,as well as the ratio of P-P38/P38.Conclusion:Electro-acupuncture on Jiaji points can down-regulate the expression of P-P38 in motor neuron of anterior horn of lumber spinal cord in the middle age of ALS-SOD1^G93A transgenic mice,and inhibit the activation of P38 MAPK to protect the neuron.
