摘 要:为了探明Ca.Brocadia型厌氧氨氧化污泥胞外聚合物(extracellular polymeric substances,EPS)自发荧光的特性与来源,采用三维荧光光谱、荧光原位杂交、高效液相色谱法、标准加入法等进行了探究。结果表明,Ca.Brocadia型厌氧氨氧化污泥的EPS主要呈现4个荧光峰,其中352/464nm和442/521nm这2个荧光峰较为特殊,其来源分别为细胞代谢物辅酶NADH和核黄素的胞外分泌,并且Ca.Brocadia型厌氧氨氧化污泥EPS中这2种荧光物质的质量摩尔浓度高于硝化污泥以及Ca.Jettenia型厌氧氨氧化污泥中2种荧光物质的质量摩尔浓度。研究结果可为Ca.Brocadia型厌氧氨氧化反应器的运行以及厌氧氨氧化菌Ca.Brocadia代谢机制的深入探究提供参考。To study the Ca. Brocadia of anammox sludge extracellular polymer (extracellular polymeric, substances, EPS) char- acteristics and the sources of spontaneous fluorescence, three-dimensional fluorescence spectroscopy and fluorescence in situ hy- bridization, HPLC, standard addition method were used to explore. Excitation-emission matrix fluorescent spectroscopy analysis show that the EPS has four main fluorescent peaks, and peaks of 352/464 nm and 442/521 nm are relative special among them. Coenzyme NADH and riboflavin are verified as the sources of those two special autofluorescences via fluorescent in situ hybridiza- tion, excitation-emission matrix fluorescent spectroscopy, HPLC and standard addition method. And it is found that fluorescent substances contents are higher in Ca. Brocadia-dominant anammox sludge than in nitrifying sludge and Ca. Jettenia-dominant an- ammox sludge. The results may provide reference for Ca. Brocadia-dominant anammox reactor operation and exploring Ca. Brocadia metabolic mechanism.
