摘  要:茶叶经甲醇回流,热水提取,Sevag法去蛋白,Sephadex G-100柱层析得纯化茶叶多糖(TP)。聚丙烯酰胺凝胶电泳证明其为均一多糖,凝胶过滤法测得其平均分子量为91 000。气相色谱分析含L-岩藻糖,D-甘露糖,D-葡萄糖,D-半乳糖和阿拉伯糖,其摩尔比为0.23:1.04:0.62:2.43:1.00。纸层析表明含葡萄糖醛酸,其含量为80%。原子吸收光谱表明TP富含多种微量金属元素。动物实验表明TP具有降血脂作用:腹腔注射25 mg/kg和50 mg/kg,正常小鼠血清总胆固醇分别下降18%和24%;口服50 mg/kg和100 mg/kg,可明显对抗小鼠实验性高胆固醇血症的形成,血清总胆固醇分别下降34%和43%;口服22.5 mg/kg×10d和45 mg/kg×10 d,可使高脂血症大鼠血清总胆固醇分别下降12%和17%,甘油三酯含量降低15%和23%;低密度脂蛋白胆固醇分别下降6%和29%,高密度脂蛋白胆固醇均增加26%。Water-soluble polysaccharide from tea leaves was isolated with hot water after reflux with methanol and further purified by gel filtration on Sephadex G-100. The purified polysaccharide (TP) was demonstrated as homogeneous by electrophoresis. The average MW of TP was found to be 91000. It was composed of fucose, mannose, glucose, galactose and arabinnose in the molar ratio of 0.23: 1. 04: 0.62: 2.43: 1.00 and also contained 80% glucuronic acid. Atomic absorption spectrum showed that TP was abundant in several trace elements which were healthful to human body. TP presented artihyperlipidemia activity in animal tests: TP (ip 25 mg/kg and 50 mg/kg) reduced cholesterol contents in serum of mice by 18% and 24%; at doses of po 50 mg/kg and 100 mg/kg, cholesterol contents in serum of mice with hypercholesterolemia decreased by 34% and 43%; TP lowered total cholesterol, triglyceride and low density lipoprotein (LDL) contents in serum of rats with hyperlipidemia by 12%, 15% and 6% at the dose of 22.5 mg/kg×10 d and by 17%, 23% and 28% at the dose of 45.0 mg/kg×10 d, while high density lipoprotein (HDL) increased by 26% at both doses.
