摘  要:目的观察姜黄素对糖尿病大鼠视网膜E26转录因子-1(Ets-1)表达的影响,探讨姜黄素治疗抑制糖尿病视网膜病变的可能机制。方法采用腹腔注射链尿佐菌素(STZ)60mg/kg诱导大鼠糖尿病模型,将20只造模成功的动物随机分成糖尿病组(10只),姜黄素治疗组(10只),取等量正常大鼠作为空白对照组。治疗组在大鼠出现糖尿病表现后按姜黄素200mg/(kg·d)灌胃,姜黄素用质量分数1%羧甲基纤维素配成混悬液后给药,共8周。糖尿病组及治疗组同期给予等量的1%羧甲基纤维素灌胃,连用8周。8周后取大鼠眼球分别行免疫组化、用ELISA法测定Ets-1浓度。结果 ELISA法测得糖尿病组的Ets-1浓度比正常组增加(P<0.05),治疗组Ets-1浓度比糖尿病组低(P<0.05),但比正常对照组高(P<0.05)。免疫组化显示正常对照组视网膜中Ets-1阳性细胞微量表达,糖尿病组及姜黄素治疗组大鼠视网膜组织中Ets-1阳性细胞大量表达,且姜黄素治疗组大鼠视网膜组织中Ets-1阳性细胞表达程度低于糖尿病组。电镜结果提示糖尿病组及姜黄素治疗组均可看到糖尿病视网膜病变的病理改变,但姜黄素治疗组的病理改变程度比糖尿病组要轻。结论姜黄素可抑制糖尿病大鼠视网膜ETS-1的表达,从而缓解其糖尿病视网膜病变进程。Objective To observe the effects of Curcumin on the expression of E26 transformation specific-1(Ets-1) in the retina of diabetic rats. Methods 20 SD rats of successful diabetic models were randomly divided into diabetes group and Curcumin treatment group,which were induced into diabetes by streptozotocin(60mg·kg-1) intraperitoneal injection each group 10 rats. Another 10 normal rats were recruited as normal control group. Cureumin 200mg/(kg·d) containing carhoxymethylcellulose was administered intragastrically last for 8 weeks after the models formed in Curcumin treatment group. And only oarboxymethylcellulose was given at the same way in diabetes group and normal control group for 8 weeks. After 8 weeks,the eyes of 10 rats in each group were removed,and used to detected the expression of Ets-1 in the retinas by ELISA and immunohisochemistry. Results The results of ELISA showed that the expression level of Ets-1 in retina in diabetes group was significantly higher than normal control group(P〈0.05). The Curcumin treatment group was lower than diabetes group(P〈0.05),but higher than control group(P〈0.05). The results of immunohisochemistry showed that the expression of Ets-1 positive cells in retina was trace expressed in control group,but strongly expressed in diabetes group and Curcumin treatment group,and especially the diabetes group. The pathological changes of diabetic retinopath were observed by electron microscopy in diabetes group and Curcumin treatment group. The degree of pathological changes in Curcumin treatment group was lower than those in diabetes group. Conclusion Curcumin could inhibits the expression of Ets-1 in retina of diabetic rats and alleviates the progression of diabetic retinopathy.
