摘 要:烟草类胡萝卜素对烟叶品质和可用性有重要作用,ζ-胡萝卜素脱氢酶(ZDS)是调控类胡萝卜素合成积累的关键酶之一。为研究ZDS基因干扰表达对烟草类胡萝卜素生物合成的影响,从贵州省烟草科学研究院福泉试验基地取样品,采用不同株系相同部位的方式取样,与贵烟1号(GY1)对照,筛选出能够构建RNA干扰(RNAi)载体并成功转化的转基因植株,建立ZDS基因的SYBR Green I实时荧光定量RT-PCR的检测方法,检测新鲜烟叶中ZDS基因表达水平并分析其遗传表型的变化。结果表明,ZDS和其他类胡萝卜素合成关键酶基因的表达均下调,其遗传后代出现异于正常植株的白化与矮化的表型。证明了ZDS基因对产生这种异同发挥了作用,也充分佐证了RNAi技术在基因功能分析中有着重要的作用,但这些样品表现出白化和矮化的表型是否是由于烟草类胡萝卜素的合成受到抑制尚不确定,还需要进一步验证。Tobacco earotenoids had an important effect on leaf quality and availability.ζ-carotene desaturase (ZDS)was a key enzyme in the regu]ation of carotenoid biosynthesis.In order to study the interference of ZOS gene expression of carotenoid biosynthesis in tobacco,this paper took samples from Fuquan test base of Guizhou Provincial Academy of Tobacco Science ,using different strains of the same parts of the sampling methods, scalding out the transformed transgenic plants of RNA interference (RNAi)vector from tobacco on the 1st (GY 1 ).ZDS gene SYBR Green I RTFQ RT- PCR test method was built for detecting fresh tobacco leaves ZDS gene expression levels and phenotypic analysis of genetic variation.The results showed that ZDS and other carotenoid synthesis gene expression of key enzymes decreased,and their genetic offspring albino and dwarf phenotype were different from normal plants. It proved that ZDS genes played a key role in those similarities and differences,and it also demonstrated that the RNAi technology played an important role in the analysis of gene function.But it was not sure that the albino and dwarf phenotype of these samples was due to carotenoid synthesis inhibited in tobacco and it needed further validation.
