Determination of p-(3,3-Dimethyl-1-Triazneo) Benzoic Acid by Capillary Gas Chromatographic Method

摘  要:将羧苯氮烯钠甲酯化,以十八烷作内标,采用毛细管气相色谱法测定了羧苯氮烯钠的小鼠血药浓度。血浆样品经酸化用醋酸乙酯提取,回收率大于95%。该方法的线性范围为2~20μg,检测限为0.1μg。采用8 m×0.3 mm I.D.的SE30石英毛细管柱,柱温155~240℃,进样器和检测器温度为260℃,检测器为FID,载气为高纯氮气,柱前压0.3 kg/cm^2,每份样品的分析时间不超过15 win。小鼠iv 10 mg/kg后,血药一时间数据用二房室模型描述,其药物动力学参数α和β分别为0.157 min^(-1)和0.0134 min^(-1)。A capillary gas chromatographic method was used to measure methyl derivative of p-(3,3-dimethyl-1-trizeno) benzoic acid (CBDT) in plasma for evaluation of its pharmacokinetics in mice. Octadecane was used as an internal standard. Mouse plasma samples were adjusted to acidic pH and subjected to a single extraction with ethyl acetate. CBDT recovery from plasma exceeds 95%. The method was linear over the range of 2.0—20 μg/ml. The working detection limit was 0.1 μg. 8 m×0.33 mm I.D. fused silica capillary column packed with SE30 was applied to analysis. Column temperature rose from 155℃ to 240℃. Injector and detector (FID) temperature was 260℃. Carrier gas was N_2 (99.999%) and was kept in 0.3 kg/cm^2 column pressure. Analysis time was under 15 min. The pharmacokinetics of CBDT in mice were fitted by an open two compartment model. The composite parameters following iv injection of 10 mg/kg were; α, 0. 157 min^(-1) and β, 0. 0134 min^(-1) respectively.
