摘 要:[目的]采用超高效液相色谱-四极杆-飞行时间质谱(UHPLC-Q-TOF/MS)研究羊耳菊提取物在大鼠循环肠灌流液中的代谢产物。[方法]收集健康SD大鼠循环肠灌流液,分别用甲酸水溶液和正丁醇处理样品后,UHPLC-Q-TOF/MS分析其代谢产物。结合对照品、质谱碎片信息和相关文献,初步推测代谢产物的结构。[结果]大鼠循环肠灌流液中的代谢物较少,初步鉴定出3个代谢产物,主要存在二咖啡酰基奎宁酸酯基位置异构和甲基化代谢物。[结论]该方法初步探究了羊耳菊提取物主要成分在大鼠循环肠灌流液中的代谢特征,为阐释羊耳菊药材的药效物质基础提供理论依据。[Objective]To investigated the metabolites of Inula cappa extract in the intestinal circulating perfusion fluid of rats by UHPLC-Q-TOF/MS.[ Method] The healthy intestinal perfusion fluid of SD rats was collected and the samples were treated with formic acid aqueous solution and n-butanol respectively.Identification and structural elucidation of the metabolites were performed by comparing the changes in molecular mas-ses,retention times and full scan metabolisms (MS) spectra with those of the parent drug,and the relative data of authentic reference.[Results] There were fewer metabolites in the intestinal circulating perfusion fluid of rats,and three metabolites were identified.There were mainly isomers and methylated metabolites of di-caffeoylquinic acid ester groups.[ Conclusion] The method is used to study the metabolic characteristics of the main components of the extract of Inula cappa extract in the intestinal circulating perfusion fluid of rats,and to provide the theoretical basis for e-lucidating the pharmacological basis of the herbs.
