高效液相色谱法测定猪肾脏中赭曲霉毒素A

摘　　要：建立高效液相色谱法测定猪肾脏中赭曲霉毒素A(OTA)的含量。采用免疫亲和柱净化,Zorbax SB-C18柱(4.6 mm×250 mm,5μm)为色谱柱,乙腈-2%乙酸为流动相,激发波长(λem)为332 nm,发射波长(λex)为460 nm。结果显示,赭曲霉毒素A在0.1 ng/mL^10 ng/mL范围内线性关系良好(R^2=0.999 9),平均加样回收率为77.3%~84.4%,平均相对标准偏差(RSD)为0.8%~1.3%。该方法简便、准确性好、灵敏度高,可用于猪肾脏中赭曲霉毒素含量的测定。The orhratoxin A （OTA） content in the kidney of pigs was determined by high performance liquid chromatography. The Zorbax SB-C18 （4.6 mmx250 mm,5μm） column was used with the mobile phase of ace-tonitrile-2%acetic acid and the detection of extraction wavelength 332 nm and emission wavelength 460 nm, and purified by immunoaffinity column. The results showed that the OTA content had good linear relationship in the range of 0.1 ng/mL-10 ng/mL（R2=0.9999）, the average recovery was 77.3%-84.4%,and relative standard deviation （RSD） was 0.8%-1.3 %.The method was simple, accurate and sensitive for the determination of ochratoxin in the kidney of pigs.
