摘　　要：目的为鼠疫F1抗体检测金标试纸条筛选出较为合适的样本稀释液,以有效避免其非特异性反应,提高试纸条的检测质量。方法选取4种血清样本稀释液,对同一份鼠疫菌免疫兔血清稀释后作鼠疫F1抗体检测,筛选出不影响敏感性的稀释液;用该稀释液检测既往鼠疫患者血清、既往发生非特异反应的血清及现场样本,验证该稀释液的检测效果。结果 4种稀释液检测同一份鼠疫菌免疫兔血清的最高检出滴度无明显差异;用含0.02 mol/L磷酸盐缓冲液(PBS)pH7.4、含0.5%牛血清白蛋白第五组份(ABV)生理盐水、含5%兔血清生理盐水对比检测10份既往鼠疫患者血清检测均为阳性;用含0.5%ABV生理盐水检测37份既往发生非特异反应血清和293份现场采集犬血清均为阴性。结论含0.5%ABV生理盐水能有效避免胶体金免疫层析法(GICA)非特异性反应的发生而不影响敏感性,可作为鼠疫F1抗体检测金标试纸条的稀释液。Objective To improve the quality of gold-immunochromatography assay for yersinia pestis F1 antibody test, and to screen a more suitable specimen diluent in order to avoid non-specific reaction effectively. Methods Screening the diluent that did not affect the sensitivity of GICA through 4 kinds of serum specimen diluent. The detection was be made to the same yersinia pestis immune rabbit serum respectively. Gold-immunochromatography assay was used to detect the effect of diluent of former plague patients serum, non-specific reaction serum and field investigation serum samples. Results The plague F1 antibody highest detection titer of the same yersinia pestis immune rabbit serum showed that there was no significant difference in 4 kinds of serum specimen diluent with GICA. 10 former plague patients serum were diluted by 0.02 mol/L PBS （pHT.4）, 0.5% ABV NS 5% rabbit serum NS respectively for detecting the plague F1 antibody were positive. The plague F1 antibody of both 37 non-specific reaction serum and 293 field investigation serum samples were negative diluted by 0.5% ABV NS. Conclusion 0.5%ABV NS can effectively avoid non-specific reaction of GICA, and does not affect the sensitivity.It can be used as serum specimen diluent of GICA for detecting the plague F1 antibody.
