摘 要:目的:探讨人胎儿侧胎盘间充质干细胞对急性肺损伤患者的炎症调节作用。方法:采用酶消化法从人胎盘间充质干细胞中分离出胎儿侧胎盘间充质干细胞,与急性肺损伤患者的外周血淋巴细胞共培养,MTT方法确定细胞增殖,ELISA方法测定炎症因子的变化,流式细胞术检测淋巴细胞亚群的变化。结果:人胎儿侧胎盘间充质干细胞能够抑制急性肺损伤患者淋巴细胞的增殖(t=3.154,P=0.008),下调γ-干扰素(interferon-γ,IFN-γ)的分泌(t=162.94,P=0.000),上调白细胞介素(interleukin,IL)-10的分泌(t=52.848,P=0.000),并下调CD4+与CD8+T淋巴细胞的比例(分别为t=15.564、P=0.004,t=7.506、P=0.017)。结论:人胎儿侧胎盘间充质干细胞能够下调急性肺损伤患者的炎症反应。Objective: To isolate and investigate the inflammation regulation function of human placenta - derived mesenchymal stem cells (hPMSCs) on acute lung injury. Methods: The enzyme directory digestion method was used to isolate hPMSCs. The hPMSCs were added to the lymphocyte of acute lung injury, with the proliferation of lymophocytes measured by MTr method, and the inflammatory cytokines detected by ELISA. The surface phenotypes of T lymphocytes were analyzed by flow cytomety before and after co -culture with hPMSCs. Results: hPMSCs suppressed the proliferation ofT cell in patients with acute lung injury ( t =3. 154, P = 0. 008 ), down - regulated the level of IFN -γ( t = 162.94, P = 0. 000 ), and up - regulated the level of IL10 ( t = 52. 848, P = 0.000). The CD4^ + T cell subsets CD8 ^+ T cell subsets were suppressed ( respectively t = 15. 564, P = 0.004; t = 7. 506, P = 0. 017). Conclusion: hPMSCs can significantly down - regulate the in- flammation of acute lung injury.
