摘 要:目的基于荧光免疫层析技术平台,针对甲型流感病毒抗原,开发甲型流感病毒快速诊断试剂。方法利用荧光微球标记甲型流感病毒单克隆抗体、生物素标记甲型流感病毒单克隆抗体以及包被亲和素和羊抗鼠IgG多克隆抗体的检测卡,构建甲型流感病毒检测体系。通过原料评价、校准曲线、空白限、准确度、线性范围、重复性、热稳定性及临床比对,验证试剂诊断性能。结果本研究得到的荧光免疫层析试剂具有准确度高、检测范围宽、重复性和热稳定性好等优点,其正确率及敏感率均明显高于经典的胶体金试剂。结论本研究应用将为国境口岸或医院的甲型流感病毒快速筛查提供方便、快捷、灵敏的技术手段。Objective To develop a rapid diagnostic kit for influenza A virus based on the platform of fluorescence immunochromatography. Methods The detection system for influenza A virus was constructed by using the fluorescent microsphere to label the influenza A virus monoclonal antibody, the biotin to label monoclonal antibody against influenza A virus, and the assaying card to detect the coated avidin and the goat-anti-mouse IgG polyclonal antibody. The diagnostic performance was validated by detecting the raw materials of the test strip, calibration curve, blank limit, accuracy, measurement range, repeatability and thermal stability and comparing the results of the clinical trials. Results The fluorescence immunochromatographic strip obtained in the study has the advantages of high accuracy, wide measurement range and good repeatability and thermal stability, and its accuracy and sensitivity are significantly higher than those of immunochromatogragphic assay. Conclusion The technology will provide a convenient, quick and sensitive diagnostic reagent for the rapid screening of influenza A virus in the frontier port or hospital.
