摘 要:目的观察参茸护肤乳对皮肤衰老的保护作用及可能作用机制。方法 71只小鼠按抽签法分为空白组、模型组、基质组、阳性对照组及参茸高、中、低剂量组各9只。除空白组外,其余各组通过皮下注射D-半乳糖建立皮肤衰老模型。造模后基质组涂抹护肤乳液基质;阳性对照组涂抹复颜滋润抗皱乳液;参茸高、中、低剂量组分别涂抹含12%人参提取物和11%鹿茸胶原多肽的护肤乳、含7%人参提取物和5%鹿茸胶原多肽的护肤乳、含3%人参提取物和2%鹿茸胶原多肽的护肤乳;各组给药剂量均为1 mg/cm^2,共给药42天。观察各组小鼠皮肤组织中的水分、透明质酸、羟脯氨酸、丙二醛(MDA)含量及谷胱甘肽过氧化氢酶(GSH-Px)、超氧化物歧化酶(SOD)活性,另外取8只小鼠进行急性多次皮肤刺激性实验观察参茸护肤乳对小鼠皮肤的刺激性。结果参茸护肤乳对皮肤安全、无刺激性。模型组小鼠皮肤中水分、透明质酸和羟脯氨酸含量及GSH-Px、SOD活力均下降,MDA含量升高(P<0.01)。小鼠皮肤经涂抹高剂量参茸护肤乳后,小鼠皮肤的水分、透明质酸和羟脯氨酸的含量及SOD、GSH-Px活性显著升高,MDA含量显著降低(P<0.05或P<0.01),且与复颜滋润抗皱乳液效果相当。结论参茸护肤乳具有明显改善皮肤衰老的作用,提高皮肤衰老小鼠的皮肤水分、透明质酸、羟脯氨酸含量及SOD、GSH-Px活性,降低MDA含量可能是其机制之一。Objective To observe the protective effect of Shenrong Body Lotion( 参茸护肤乳,SBL) on skin aging and its potential mechanism. Methods By drawing lots,71 mice were divided into a blank group,a model group,a matrix group,a positive control group and Shenrong high,medium and low dose groups,with 9 mice in each group. D-galactose was injected subcutaneously to establish the skin aging model in all groups except for the blank group. After modeling,the matrix group was applied the body lotion matrix for skin care,the positive control group was given Revitalift Anti-Wrinkle Emulsion,while the Shenrong high,medium and low dose groups were respectively given body lotion containing 12% Renshen [人参 Ginseng Radix Et Rhizoma] extract and 11% Lurong [鹿茸 Cervi Cornu Pantotrichum] collagen peptide,7% Renshen extract and 5% Lurong collagen peptide,and 3% Renshen extract and 2% Lurong collagen peptide. The dose was 1 mg/cm^2 for all groups,and the treatment period was 42 days. The contents of water,hyaluronic acid,hydroxyproline,and malondialdehyde( MDA),and the activity of glutathione peroxidase( GSH-Px) and superoxide dismutase( SOD) in the skin tissues were observed. There were also another 8 mice subjected to acute multiple skin irritation test to observe the irritation of SBL for the skin of mice.Results SBL was safe and non-irritating for skin. The content of water,hyaluronic acid and hydroxyproline decreased,and the activity of GSH-Px and SOD also decreased while the content of MDA increased in the model group( P〈0. 01). After the application of high dose SBL,the contents of water,hyaluronic acid and hydroxyproline and the activity of SOD and GSH-Px in the skin significantly increased while the content of MDA significantly decreased( P〈0. 05 or P〈0. 01),and the effect was quite similar to that of Revitalift Anti-Wrinkle Emulsion.Conclusion SBL seems to have significant effect on improving skin aging,and the potential mechanism might be improving the content of skin moisture,hyalur
