摘  要:目的:观察人绒毛膜促性腺激素(hCG)对K9小鼠睾丸间质细胞(K9细胞)转醛醇酶(transaldolase,TAL)活性的影响,探索哺乳动物中TAL活性与睾酮合成之间的关系。方法:酶联免疫法测定细胞培养液中睾酮分泌量,TAL活性测定,蛋白免疫印迹(Westernblotting)分析TAL蛋白变化及RNA印迹法(Northenblotting)分析TALmRNA表达情况。结果:hCG实验组睾酮分泌量及TAL活性比对照组均提高2倍,Westernblotting分析TAL蛋白量无明显变化,而Northenblotting分析TALmRNA表达明显增加。结论:hCG促进K9细胞中睾酮合成的同时TAL活性显著增加,TAL可能与哺乳类K9细胞类固醇激素的合成有关。Objective:To study effects of human chorionic gonadotropin (hCG) on the activity of transaldolase(TAL)and probe the relationship between TAL activity and biosynthesis of testosterone in mammals.Methods:The testosterone in the medium of cell incubation was measured by EIA,the activity of TAL was measured by the method described by Lachaise,the quantity of TAL proteins was detected with the method of Western Blotting and the expression of TAL mRNA with the method of Northern Blotting.Results:There was a 2 fold increase in both TAL activity and synthesis of testosterone in hCG experimental group, and no alteration in quantity of TAL proteins by Western Blotting analysis. But the expression of TAL mRNA was obviously increased by Northern Blotting analysis.Conclusion:hCG may enhance the synthesis of testosterone and TAL activity. Therefore, there may be a relationship between the activity of TAL and the synthesis of testosterone in mammal′s Leydig cells.
