摘  要:目的:研究^(131)I-Herceptin在胃癌裸鼠模型体内的生物学分布及^(131)I-Herceptin的抑瘤效果。方法:采用Iodogen法制备^(131)I-Herceptin。以NCI-N87胃癌细胞皮下接种BALB/c-neu裸鼠建立动物模型,对荷瘤小鼠模型进行SPECT连续显像。测量小鼠用药后的4、12、24、48 h各脏器每克组织每分钟的放射性计数(epm/g),并计算T/NT以及每克组织的放射性计数占注射剂量放射性计数的百分比(%r D/s)。将荷瘤裸鼠随机分为3组:^(131)I-Herceptin组、Herceptin组与生理盐水对照组。接种前后测量肿瘤长短径,并计算肿瘤生长的抑制率,流式细胞仪检测肿瘤细胞周期及细胞凋亡率。结果:实验组对比对照组,显像对比明显;实验组T/NT以及肿瘤组织%ID/g显著高于对照组(P<0.01)。3组间肿瘤抑制率及凋亡率均差别明显(F分别为84.59、304.38,P<0.0001),其肿瘤抑制率及凋亡率由高到低分别为^(131)I-Herceptin组、Herceptin组与生理盐水组。结论:^(131)I-Herceptin在胃癌裸鼠肿瘤组织中有良好的靶向作用;^(131)I-Herceptin具有良好的抑瘤作用。Objective: To observe the biodistribution and anticancer effect of ^(131)I-Herceptin in nude mouse model of gastric cancer. Methods: Herceptin was labelled with ^(131)I by an Iodogen method. The animal model was made by implanting NCI-N87 cells subcutaneously. Tumor bearing mice were imaged continuously with SPECT. Radiocountings per minute were recorded at 4, 12, 24 and 48 h after the mice were injected with the drugs or normal saline(NS) for calculating the T/NT and the uptake percentage per gram of tissue to the injected dosage. The tumor-burdened nude mice were divided into 3 groups randomly: ^(131)IHerceptin group, Herceptin group and NS group. The length and width of inoculated tumor were measured before and after treatment to calculate the tumor growth inhibition rate. The tumor cell cycle and cell apoptosis rate were detected by flow cytometry. Results: Compared with the control group, the imagination of the experimental group was obvious. The values of T/NT and tumor tissue % ID/g of the experimental group were higher than those of the control group(P 0.01). The differences of the inhibition rates and apoptosis rates among the three groups were significant,(F=84.59, 304.38, P0.0001) with the high to low order as the ^(131)I-Herceptingroup, Herceptin group and NS group. Conclusion: ^(131)I-Herceptin has excellent targeting and inhibitory effects on gastric cancer in nude mice tissues.
