摘 要:近年血清4型禽腺病毒(Serotype 4 fowl adenovirus,FAdV-4)在国内鸡群流行,给养鸡业带来了巨大经济损失。目前尚无针对FAdV-4特异性单克隆抗体的制备及其应用报道。研究通过将FAdV-4全病毒免疫小鼠,以纤突蛋白2原核表达产物为筛选抗原,研制出了4株针对FAdV-4的单克隆抗体,分别命名为3C2、4A6、5C4、5H6。间接免疫荧光试验证明4株单克隆抗体不与所检测的血清8型禽腺病毒反应,也不与FAdV-4的纤突蛋白1反应,仅与FAdV-4的纤突蛋白2反应,并且单克隆抗体3C2在体外能有效抑制FAdV-4的感染。4株抗FAdV-4纤突蛋白2单克隆抗体的成功获得为研制FAdV-4快速诊断试剂盒及探究纤突蛋白2在FAdV-4致病机制中的作用奠定了基础。In recent years, the serotype 4 Fowl adenovirus (FAdV-4)has been prevalent in the chicken flocks in China, which has caused large economic losses to the poultry industry. However, there were no reports on the preparation and application of specific monoclonal antibody against FAdV-4. In this study, four monoclonal antibodies against FAdV-4 screened by prokaryotic expression products of fiber2 protein were developed by immunizing mice with whole virus of FAdV-4, which were designated as 3C2, 4A3, 5C4 and 5H6, respectively. Indirect immunofluorescence assay showed that these antibodies neither reacted with serotype 8 fowl adenovirus, nor reacted with FAdV-4 fiberl protein, and only reacted with FAdV-4 fiber2 protein. In addition, the infection and replication of FAdV-4 could be effectively inhibited by monoclonal antibody 3C2 in vitro. The preparation of the four monoclonal antibodies against FAdV-4 fiber2 protein laid the foundation for further establishment of the rapid diagnostic kit for FAdV-4 and the study on the role of fiber2 protein in pathogenesis of FAdV-4.
