摘 要:目的:研究黄连对小鼠急性肝毒性。方法:通过黄连提取物高剂量组(0.5 mg/m L)、中剂量组(0.025 mg/m L)、低剂量组(0.012 5 mg/m L)灌胃给药,观察其对小鼠临床症状及病理变化的影响。结果:不同剂量组黄连对小鼠肝脏造成不同程度的损伤,其中高剂量组(0.5 mg/m L)小鼠肝脏显著肿胀,呈暗黑色,显微镜观察病变明显,H.E染色肝脏中度脂肪变性;中剂量组(0.025 mg/m L)肝脏呈暗红色,肝脏轻微脂肪变性;低剂量组(0.012 5 mg/m L)肝脏呈暗红色,显微镜下观察可见炎症细胞浸润。结论:黄连提取物高、中、低剂量组均能引起小鼠不同程度的肝脏损伤,损伤程度随黄连剂量增加而加重。Objective: To investigated the toxicoty of various doses of Rhizoma Coptidis on hepatocytes of mice and guide safe medication. Methods: High(0.5 mg/m L),middle(0.025 mg/m L)and low(0.012 5 mg/m L)dose of Rhizoma Coptidis were intragastric administrated to mice in each group. General conditions and the results of H.E staining were recorded. Results: General conditions of mice became deteriorated with the increasing of Rhizoma Coptidis dose. The liver biopsy pathological changes of mice were increased with the increasing of Rhizoma Coptidis dose. Statistically significant differences were detected among all groups except between the normal control and the low-dose group. The liver of mice from the low-dose group showed fat degeneration at the beginning,and it worsened along with the increasing of Rhizoma Coptidis dose. Conclusion: Rhizoma Coptidis can cause liverdamage in mice at varying degrees,and the effect will become stronger with the increasing of Rhizoma Coptidis dose.
