摘  要:目的探索使用PEI-Fe_3O_4纳米颗粒标记人羊膜上皮细胞(h AEC)。方法采用透射电子显微镜和动态光散射表征PEI-Fe_3O_4纳米颗粒。使用纳米颗粒标记原代培养的h AEC,普鲁士蓝染色评价其标记效率,台盼蓝染色法和CCK-8法检测细胞存活率和细胞毒性。结果 PEI-Fe_3O_4纳米颗粒呈紧凑的球形,平均粒径为13 nm,水动力学半径为17.56 nm,zeta电位为+34.5 m V。浓度大于20μg/ml的纳米颗粒对h AEC的标记效率达到91%。50(t=16.37,P<0.0001;t=10.39,P<0.0001)、100μg/ml(t=29.89,P<0.0001;t=16.86,P<0.0001)纳米颗粒浓度下的h AEC存活率和细胞活力均明显低于未标记时。结论 PEI-Fe_3O_4纳米颗粒可用于标记h AEC,在其工作浓度下未表现出显著的细胞毒性。Objective To label human amniotic epithelial cells( h AECs) by using PEI-Fe_3O_4 nanoparticles. Methods The PEI-Fe_3O_4 nanoparticles were characterized by using transmission electron microscopy and dynamic light scattering. The primary cultured h AECs were labeled with the nanoparticles,and the labeling efficiency was evaluated by Prussian blue staining. The cell survival rate and viability were tested by using placenta blue staining and CCK-8 assay,respectively. Results The PEI-Fe_3O_4 nanoparticles were compact spheres with an average particle size of 13 nm,a hydrodynamic radius of 17. 56 nm,and a zeta potential of + 34. 5 m V.The labeling efficiency of the nanoparticles on h AECs reached 91% when the concentrations were greater than20 μg/ml. When the concentrations of nanoparticles were at 50 μg/ml( t = 16. 37,P < 0. 0001; t = 10. 39,P < 0. 0001) and 100 μg/ml( t = 29. 89,P < 0. 0001; t = 16. 86,P < 0. 0001),the cell survival rates and cell viabilities were significantly reduced versus controls. Conclusion The PEI-Fe_3O_4 nanoparticles can be used for labeling h AECs without obvious cytotoxicity at its working concentration.
