过表达GCNF诱导骨髓间充质干细胞凋亡的相关研究

摘　　要：目的研究过表达生殖细胞核因子(GCNF)诱导骨髓间充质干细胞(BMSCs)凋亡的可能机制。方法通过过表达GCNF腺病毒上调BMSCs中GCNF表达水平,利用间接免疫荧光化学技术确定其亚细胞定位,MTT检测细胞活力的影响,DAPI染色检测细胞凋亡,Caspase3/8/9检测细胞凋亡蛋白活性。结果 BMSCs细胞中GCNF过表达后,GCNF蛋白亚细胞定位于细胞核,细胞活力降低,凋亡通路被激活。结论腺病毒GCNF过表达能降低大鼠BMSCs细胞活力,增加其凋亡水平。Objective To study the effect and mechanism of apoptosis of bone marrow mesenchymal stem cells induced by overexpression of germ cell nuclear factor(GCNF).Methods GCNF expression was upregulated in BMSCs by overexpression of GCNF adenovirus,and subcellular localization was determined by indirect immunofluorescence technique,cell viability was detected by MTT,and DAPI staining was used to detect cell apoptosis,cell apoptosis protein activity was detected by Caspase3/8/9.Results After overexpression of GCNF in BMSCs cells,the subcellular localization of GCNF protein was found in the nucleus,the cell viability was decreased and the apoptotic pathway was activated.Conclusion Overexpression of adenovirus GCNF can decrease the activity of BMSCs cells and increase the level of apoptosis.
