调神通络针刺法对缺血再灌注大鼠海马区神经元形态及Cx43半通道影响的实验研究

摘　　要：目的:探讨调神通络针刺法对缺血再灌注大鼠海马区神经元形态的影响及对Cx43半通道的相关作用机制。方法:以经脑缺血再灌注处理的健康雄性Wistar大鼠为研究对象,分为正常组、假手术组、模型组、针刺组、非穴位针刺组,以针刺治疗为干预手段,采用细胞HE染色法镜下观察大鼠海马区的神经元形态结构及病理改变;用荧光定量PCR检测大鼠海马区Cx43、m GluR5、P38MAPK mRNA含量表达。结果:正常组与假手术组Cx43、m GluR5、P38MAPK mRNA含量及神经细胞形态结构无差异;模型组Cx43、m GluR5、P38MAPK mRNA含量明显高于正常组(P<0.05),神经细胞损伤明显,提示造模成功;针刺组Cx43、m GluR5、P38MAPK mRNA含量较模型组低(P<0.05),病理改变与正常组相同;非穴位针刺组Cx43、m GluR5、P38MAPK mRNA含量及病理改变与模型组无差异。结论:调神通络针刺法能够有效抑制Cx43、m GluR5、P38MAPK mRNA的表达,调控Cx43磷酸化和去磷酸化过程,进而调节Cx43半通道的通透性,使该通道起到最佳的脑保护作用,抑制神经细胞进一步损伤,并促进已损伤神经的恢复。Objective ： To investigate the effect of mentality - regulating and collateral - dredging acupuncture on hipp- ocampal neuron morphology and Cx43 hemichannel in rats with ischemia/reperfusion and related mechanism of action. Methods：Healthy male Wistar rats treated by cerebral ischemia/reperfusion were selected and divided into normal group, sham - operation group, model group, acupuncture group, and non - acupuncture group. Acupuncture was used as inter- vention. HE staining was used to observe the morphological structure and pathological changes of neurons in the hippocampus under a microscope, and quantitative real - time PCR was used to measure the mRNA expression of Cx43, mGluRS, and P38MAPK in the hippocampus. Results ：There were no significant differences in the mRNA expression of Cx43 ,mGluR5 ,and P38MAPK and morphological structure of neural cells between the normal group and the sham - operation group. The model group had significantly highe＇r mRNA expression of Cx43, mGluR5, and P38MAPK than the normal group （P 〈 0. 05）, and the model group had marked injury in neural cells, suggesting that the model was established successfully. The acupuncture group had significantly lower mRNA expression of Cx43, mGluR5, and P38MAPK than the model group （P 〈 0. 05 ）and similar path- ological changes as the normal group. There were no significant differences in the mRNA expression of Cx43, mGluR5, and P38MAPK and pathological changes between the non -acupuncture group and the model group. Conclusion ：Mentality -regn- lating and collateral -dredging acupuncture can effectively inhibit the mRNA expression of Cx43, mGluR5, and P38MAPK,regulate phosphorylation and dephosphorylation of Cx43 and the permeability of Cx43 hemichannel,restore the optimal cere- bral protective effect of Cx43 hemiehannel,inhibit further injury of neural cells, and promote the recovery of injured nerves.
