摘  要:目的:建立清咽爽喉方的质量标准。采用薄层色谱法(TLC)对清咽爽喉方中的麦冬、射干进行定性鉴别,以高效液相色谱法(HPLC)对菊花中的有效成分绿原酸、木犀草苷进行含量测定。方法:色谱柱:用十八烷基硅烷键合硅胶为填充剂C18(4.6mm×250mm,5μm);以乙腈为流动相A,以0.1%磷酸溶液为流动相B;检测波长为348nm。流速:1.0m L/min;柱温25℃。结果:清咽爽喉方的麦冬、射干TCL中,在与对照药材色谱相应的位置上,显相同颜色的斑点;绿原酸在7.5~750μg范围内线性关系良好,R=0.9998,木犀草苷在5.35~535μg范围内线性关系良好,R=0.9999,平均加样回收率分别为96.69%、98.47%,RSD分别为0.99%、1.62%。结论:本方法准确可行,可作为本品的质量控制。Objective:To investigate the quality standard for Qingyan Shuanghou prescription,to perform qualitative i- dentification of Radix Ophiopogonis and blackberry lily rhizonle in Qingyan Shuanghou prescription by thin layer chroma- tography(TLC) , and to determine the content of effective constituents chlorogenic acid and cynaroside in chrysanthemum by high - performance liquid chromatography ( HPLC ). Methods : HPLC was performed on a C18 column with octadecylsilane chemically bonded silica gel as the filler(4.6mm ×250mm,5μm)with a mobile phase of acetonitrile (A) -0. 1% phos- phoric acid solution (B)at a detection wavelength of 348 nm, a flow rate of 1.0mL/min, and a column temperature of 25℃. Results:TLC of Radix Ophiopogonis and blackben^y lily rhizome in Qingyan Shuanghou prescription obtained the spots with the same color at the same positions as TLC of the control medicinal material. Chlorogenic acid showed a good linear relationship within the range of 7.5 -750μg( r = 0. 9998 ) ,with an average recovery rate of 96.69% (relative stand- ard deviation [ RSD~ = 0. 99% ). Cynaroside showed a good linear relationship within the range of 5.35 -535μg( r = 0. 9999 ) , with an average recovery rate of 98.47% (RSD = 1.62% ). Conclusion:This method is accurate and feasible and can be used for quality control of this product.
