摘  要:目的观察并比较苯巴比妥钠(PB)单独诱导、苯巴比妥钠与β-萘黄酮(β-NF)联合诱导两种方法对大鼠肝微粒体细胞色素P450(CYP450)酶含量的影响。方法将24只健康成年SD雄性大鼠随机为单独诱导组、联合诱导组及对照组,分别腹腔注射PB、PB+β-NF和生理盐水,给药4d后制备大鼠肝微粒体,测定肝脏脏器系数、肝微粒体蛋白含量、CYP450及细胞色素b5(Cytb5)含量。结果单独诱导组、联合诱导组分别与对照组比较,大鼠肝脏脏器系数、肝微粒体蛋白含量、CYP450及Cytb5含量均有增加,差异有统计学意义(P<0.05);单独诱导组和联合诱导组比较差异无统计学意义(P>0.05)。结论 PB单独作用、PB与β-NF联合作用对大鼠肝微粒体CYP450酶均产生诱导效应,两种诱导法对肝粒体CYP450酶的作用差异不明显。Objective To observe and compare the contents of liver microsomal cytochrome P450(CYP450)and b5 (Cytb5) which were induced by single and combined phenobarbital andβ-naphthoflavone in rats.Methods Totally 24 healthy adult male SD rats were randomly divided into separate induction group,combined induction group and control group.All rats in three groups were respectively intraperitoneal injection of Phenobarbital,Phenobarbital andβ-naphthoflavone,and normal saline for 4 days.Then the liver microsomes were prepared and liver/body weight ratios were measured,and the contents of protein and total CYP450 and Cytb5 of liver microsome were detected by spectrophotometry.Results Compared with the contro1 group,the liver organ coefficient,liver microsomal protein contents,CYP450 and Cytb5 contents in the separate induction group and combined induction group were significantly increased(P〈0.05).But there was no significant difference in the levels of these detection index between two experimental groups(P〉0.05).Conclusion The separate and combined effects of phenobarbital and/orβ-naphthoflavone can induce CYP450 of liver microsomes in rats,but the induction wasn't obvious difference between two methods.
