摘 要:目的研究辣椒素受体(TRPV1)在皮肤淀粉样变皮损中的表达,探讨其在皮肤淀粉样变疾病中的作用。方法选取22例皮肤淀粉样变皮损及16例正常皮肤组织,用半定量RT-PCR和免疫组化的方法研究皮损中辣椒素受体的表达情况。结果皮肤淀粉样变皮损中TRPV1的基因表达水平显著高于正常皮肤(P<0.05),免疫组化研究显示TRPV1在正常皮肤、皮肤淀粉样变的表皮角质形成细胞、附属器(毛囊、皮脂腺和汗腺)及血管内皮细胞均存在表达;TRPV1在皮肤淀粉样变皮损的表达高于正常皮肤(P<0.05)。结论 TRPV1在皮肤淀粉样变中的表达高于正常皮肤,提示其在皮肤淀粉样变致瘙痒的发生及发展中可能起着一定的作用。Objective To investigate the expression of Transient Receptor Potential Vanilloid 1(TRPV1)in cutaneous amyloidosis, and elucidate the possible role of TRPV1 in the pathogenesis of itch in the disease.Methods Tissue specimens were obtained from skin biopsy from 22 patients with cutaneous amyloidosis, and 16 from normal skins.The expression of TRPV1 was studied by RT-PCR and immunohistochemistry.Results The mRNA expression of TRPV1 was high in cutaneous amyloidosis than the normal skin by RT-PCR.The immunohistochemistry revealed that all of normal skin, cutaneous amyloidosis specimens express TRPV1 in keratinocytes, skin appendages(include hair follicles, sebaceous glands, eccrine and apocrine)and vascular endothelium cell.TRPV1 is strongly expressed in whole epidermis of cutaneous amyloidosis.There's a significant statistical difference between cutaneous amyloidosis and normal skin(P〈0.05).Conclusion The expression of TRPV1 was high in cutaneous amyloidosis than normal skin specimens, and these suggested us that TRPV1 might contribute to onset of pruritis in cutaneous amyloidosis.The role and mechanisms of TRPV1 in the pathogenesis of cutaneous amyloidosis should be established by more researchings.
