摘  要:目的观察青光安Ⅱ号方对慢性高眼压模型大鼠视网膜GSK-3β及β-catenin mRNA表达影响。方法将40只(80眼)雄性SD大鼠随机分成6组,分别为:空白组(A),模型组(B)、青光安Ⅱ号方低剂量组(C)、青光安Ⅱ号方中剂量组(D)、青光安Ⅱ号方高剂量组(E)、益脉康分散片组(F)。将B、C、D、E、F组大鼠采用烧灼巩膜表浅静脉法建立大鼠慢性高眼压模型。模型大鼠维持高眼压状态2月后开始干预,干预后2周、4周分别用q PCR检测大鼠视网膜GSK-3β及β-catenin mRNA的相对表达量。结果 2周后,与B组比较,各组GSK-3βmRNA表达降低而β-catenin mRNA表达升高,差异有统计学意义(P<0.05)。4周后,与F组比较,C、D、E组GSK-3βmRNA降低,β-catenin mRNA表达升高,差异有统计学意义(P<0.05)。与E组比较,C、D组GSK-3βmRNA降低而β-catenin mRNA表达升高,差异有统计学意义(P<0.05)。结论青光安Ⅱ号方及益脉康分散片均能抑制GSK-3βmRNA的表达,并增加β-catenin mRNA的相对表达量,对慢性高眼压大鼠视网膜神经节细胞有一定的保护作用;初步观察表明复方中药青光安Ⅱ号方在对慢性高眼压大鼠视神经的保护中,效果优于益脉康分散片,且以青光安Ⅱ号方高剂量最优。Objective To observe the effect of Qingguangan II decoction on expression of GSK-3β mRNA and β-catenin mRNA of retinal ganglion cells in chronic high intraocular pressure model rats. Methods 40(80 eyes) male SD rats were randomly divided into 6 groups, that is: blank group(A), model group(B), Qingguangan II low dose group(C), Qingguangan II medium dose group(D), Qingguangan II high dose group(E), Yimaikang dispersible tablets group(F). The rats in group B, C, D, E, F were established the model of chronic high intraocular pressure by the method of cauterizing of superficial scleral vein. Tissues of eyes were obtained after intragastric administration for 2 weeks and 4 weeks. The relative expression of GSK-3β mRNA and β-catenin mRNA was investigated by q PCR. Results At the time-point of 2-week, GSK-3β mRNA and β-catenin mRNA in group B were statistically expressed comparing with other groups(P〈0.05). Moreover, at the time-point of 4-week, GSK-3β mRNA and β-catenin mRNA in group C, D and E were statistically expressed comparingwith group F(P〈0.05). Besides, GSK-3β mRNA and β-catenin mRNA in group C and D were statistically expressed comparing with group E(P〈0.05). Conclusion Qingguangan II decoction and Yimaikang dispersion tablets can effectively inhibit the expression of GSK-3β and increase the expression of β-catenin to protect the optic nerve of rats with chronic high intraocular pressure. The experiment preliminarily indicated that, in the protection of optic nerve of rats with chronic high intraocular pressure, the high dose of Qingguang'an II was the better choice.
