摘  要:为了测定设施甜樱桃果实中的赤霉素含量,建立了甜樱桃果实中赤霉酸的高效液相色谱-串联质谱(HPLC-MS/MS)定性定量检测方法。样品经乙腈提取,液-液分配法进行净化,在SIM模式下对目标物质进行定性和定量检测,同时对设施甜樱桃激素坐果果实中赤霉酸含量进行测定。结果表明,赤霉酸的定量限为4.03μg·kg-1,在5.0~100.0μg·L-1范围内呈现良好的线性关系,相关系数R为0.9996。方法的加标回收率为87.64%~91.75%,相对标准偏差为8.9%~13.5%,5份设施甜樱桃样品中赤霉酸含量在21.3~71.1μg·kg-1。本试验建立的方法操作简单,灵敏度和重复性好,可用于甜樱桃果实中赤霉酸含量检测。A qualitative and quantitative determination method for gibberellin content in sweet cherry fruit was developed by high performance liquid chromatography-tandem mass spectrometry( HPLC-MS/MS). The sample was extracted by acetonitrile,and purified by liquid-liquid distribution method,then the sample solution was qualitatively and quantitatively detected by HPLC-MS/MS with SIM mode. The results showed that the quantification limit for gibberellin was 4. 03 μg·kg^-1,the gibberellin standard showed a good linearity in the range of 5. 0 - 100. 0 μg·L^-1 with the correlation coefficient of 0. 9996. The average recovery of gibberellin was 87. 64% - 91. 75%,and the relative standard deviations was 8. 9% - 13. 5%. The gibberellin contents in 5 samples were 21. 3 - 71. 1 μg·kg^-1. This method was simple to operate with high sensitivity and good reproducibility. It could be used to determinate the gibberellin content in sweet cherry fruit.
