摘  要:为了解近期我国华北地区猪繁殖与呼吸综合征病毒(PRRSV)的流行及变异情况,本研究对该地区某猪场采集33份2016年~2017年疑似患PRRS猪病料样品进行PCR检测,对阳性样品进行病毒分离,并从分离株中选择两株病毒(SD158和SD192)进行全基因的序列扩增及分析。结果显示,检测到13份PRRSV阳性病料样品,病原为经典美洲型PRRSV,但仅分离到11株病毒(4株感染MARC-145细胞,另外7株感染PAM细胞)。SD158和SD192株与Resp PRRS MLV株(疫苗株)的核苷酸同源性最高,分别为99.7%和99.4%。对这两株PRRSV与VR-2332、Resp PRRS MLV的各基因的推导氨基酸序列进行分析,结果显示在VR-2332与Resp PRRS MLV株22处差异氨基酸中,SD158株有15处、SD192株有14处氨基酸和Resp PRRS MLV株相同,但二者仅各有6处及7处氨基酸与VR-2332株一致,在鉴别PRRSV VR-2332株与Resp PRRS MLV株的7处可能与毒力相关的氨基酸位点时发现,SD158与SD192株各有5处基酸与VR-2332株相同。根据以上结果推测SD158与SD192株可能来源于PRRSV Resp PRRS MLV株,具有上述分子变化特征的PRRSV在我国的流行状况值得关注。To investigate the prevalence and variationof porcine reproductive and respiratory syndrome virus (PRRSV) in North China, 33 clinical samples for PRRSV detection were collected from the suspected PRRSV-infected pigs in a pig farm of North China during 2016 to 2017. The results showed that 13 samples were positive for classical North American type-PRRSV, but only 11 strains were isolated (4 strains could infect MARC-145 cells and 7 could infect PAM cells). The complete genomes of two isolates (SD158 and SD192) were sequenced, and had the highest genome nucleotide homology with RespPRRS MLV, which was 99.7% and 99.4%, respectively. There were 22 amino acid residues differences between RespPRRS MLV and VR-2332, of which 15 and 14 amino acid residues were consistent with RespPRRS MLV in SD158 and SD192 strains, respectively. There were 6 and 7 amino acid residues which were consistent with VR-2332 in SD158 and SD192 strains, respectively. In addition, seven amino acids related to virulence were identified in previous studies, while five of them in SD158 and SD192 isolates were consistent with VR-2332. Collectively, these data indicated that SD158 and SD192 were likely to be derived from RespPRRS MLV. The prevalence of PRRSV with above molecular characteristics in China is worthy of attention.
