摘 要:本试验通过建立组织笼感染模型,研究马波沙星在多杀性巴氏杆菌感染猪组织液中的药代动力学。向组织笼中注射0.5 mL对数期多杀性巴氏杆菌(5×10~8CFU/mL),感染后第24小时,对猪按体重单剂量肌内注射1.0 mg/kg和2.5 mg/kg马波沙星,按预定的时间点采集组织液样品,采用高效液相色谱(HPLC)测定组织液中马波沙星的浓度。用Win Nonlin5.2.1软件提供的非房室模型处理组织液药物浓度-时间数据。结果,不同剂量给药的主要药物动力学参数如下:1 mg/kg剂量给药:t_(max)=4 h±1.41 h,C_(max)=0.30 g/mL±0.06g/mL,AUC=9.96 g/mL·h±1.54 g/mL·h,t_(1/2β)=22.92 h±6.83 h;2.5 mg/kg剂量给药:t_(max)=5 h±1.41 h,C_(max)=0.71 g/mL±0.09 g/mL,AUC=21.06 g/mL·h±3.69 g/mL·h,t_(1/2β)=20.91 h±8.57 h。结果表明,马波沙星在多杀性巴氏杆菌感染猪组织液中的峰浓度较高,达峰时间较慢,消除半衰期较长。To study the pharmacokinetics of marbofloxacin in the tissue cage fluid of pigs infected with Pasteurella multocida,a tissue cage infection model was included in this study.Two dosages(1.0 mg/kg and 2.5 mg/kg) of marbofloxacin were administrated intramuscularly after 0.5 mL of P. multocida saline suspension was injected into tissue-cages for 24 h,and tissue cage fluid samples were sampled as designed scheme.Marbofloxacin concerntration in tissue cage fluid samples was determined by the HPLC.Concentration-time data were analysised with the non-compartment model offered by Win Nonlin software.In result,the main pharmacokinetic parameters acquired from different dosages were described as below,1 mg/kg:t_(max)=4 h±1.41 h,C_(max)=0.30 g/mL±0.06 g/mL,AUC=9.96 g/mL·h±1.54 g/mL·h,t_(1/2β)=22.92 h±6.83 h;2.5 mg/kg:t_(max)=5 h±1.41 h,C_(max)=0.71 g/mL±0.09 g/mL,AUC=21.06 g/mL·h±3.69 g/mL·h,t_(1/2β)=20.91 h ±8.57 h. In conclusion,marbofloxacin is absorbed and eliminated slowly in tissue cage fluid of pigs infected with P.multocida,and can reach high peak concentration.
