西藏绒山羊传染性脓疱的诊断与重要器官的病理学观察

摘　　要：本研究对尼玛县某羊场白绒山羊疑似患传染性脓疱进行了RT-PCR检测,并对6例病山羊进行病理剖检和病理组织学观察,以探讨其病理变化特点。PCR检测结果表明,6份样品中均扩增出579 bp的目的基因片段;对其测序分析结果表明,该序列与羊传染性脓疱病毒(CEV)基因序列同源性较高。病理剖检显示,6例病山羊程度不等口唇部、面部肿胀,口腔、舌头黏膜面有大小不等丘疹、溃疡和脓疱;瘤胃与网胃黏膜也有丘疹或结节状突起,甚至密布瘤胃黏膜表面;心脏上有大小不等坏死病灶。病理组织学检查显示,病变口腔黏膜与瘤胃黏膜上皮细胞大量增生,增生的黏膜上皮又程度不等水泡变性、气球样变,变性上皮细胞内可见嗜酸性胞浆包涵体;黏膜固有层充血、水肿,有大量单核细胞、中性粒细胞浸润,甚至形成大小不等的微脓肿;心脏均呈程度不等的变质性心肌炎或化脓性心肌炎;肺脏呈程度不等的间质性肺炎;肝、脾有程度不等的淤血、变性;全身淋巴结呈急性浆液性淋巴结炎;研究证明,除口腔、舌头外,瘤胃黏膜的病变和心脏的变质性炎症也是羊传染性脓疱病具有代表性的变化之一。以上结果证实,6只白绒山羊所患的疫病是羊传染性脓疱(羊口疮)。This experiment was conducted to confirm the contagious ecthyma of cashmere goats and explore the characteristics of the pathological changes.On the basis of clinical diagnosis,the six doubtful goats with contagious ecthyma were definitely diagnosed by RT-PCR detection technology.The pathological changes were observed by macroscopy and light microscope and special staining technique.The results showed that the specific target gene fragment 579 bp of CEV was amplified and the contagious ecthyma virus（CEV）was definitely diagnosed as the pathogen of the six sick goats.The pathological changes of autopsy showed that the lips and facials of the six sick goats were swelling.There were a large number of papules,ulcers and pustules on the mucosa of tongue and oral cavity.There were also a large number of papules on the mucosa of rumen and reticulum.The hearts had various degrees necrosis and heart failure.The histopathological changes indicated that the mucosal epithelial cells of oral cavity and rumen were hyperplasia obviously and vacuolar degeneration or balloning degeneration,and there were eosinophilic inclusion in the cytoplasm.The lamina propria of oral cavity and rumen were hyperemia and edema,and there were many monocytes and neutrophilic granulocytes and even many microabscess were formed.The hearts had various degrees parenchymatous myocarditis and suppurative myocarditis.The lungs were various degrees interstitial pneumonia.The livers and spleens were venous hyperemia and various degrees degeneration.The lymph nodes had acute lymphadenitis.Those results confirmed the important pathological changes of the mucosa of rumen and the heart are also one of the representative changes in the contagious ecthyma in goats.
