摘  要:目的研究银杏叶提取物(EGb)对大鼠急性脊髓损伤后运动功能的恢复及脊髓Nogo-A蛋白表达的影响。方法选用健康的成年Sprague-Dawley(SD)大鼠72只,雌雄不拘,体质量250~300 g。将大鼠分为正常组、急性脊髓损伤组和急性脊髓损伤后EGb治疗组(简称EGb治疗组)。采用Allen’s打击方法损伤大鼠胸椎T_8~T_(10)节段,分别于术后3、7、21 d进行BBB评分和斜板实验,并取受损节段大鼠脊髓,运用Western blot方法测定各时相点Nogo-A蛋白表达量及其变化。结果术后21 d EGb治疗组比急性脊髓损伤组BBB评分和斜板实验临界角度升高,差异有统计学意义(P<0.05)。Nogo-A蛋白在各组大鼠脊髓组织中均呈阳性表达,急性脊髓损伤组与EGb治疗组各个时相点Nogo-A蛋白表达量均显著高于正常组(P<0.05),7 d时最高,21 d有所下降,但表达量仍高于正常组。EGb治疗组在各个时间点的NogoA表达量显著低于急性脊髓损伤组,差异有统计学意义(P<0.05)。结论 EGb可促进急性脊髓损伤后大鼠运动功能的恢复,对脊髓损伤后Nogo-A蛋白表达量的升高具有明显的抑制作用。Objective To observe therapeutic effect of Ginkgo biloba extract(EGb) on motor function restoration and Nogo-A protein expression of rats after spinal cord injury(SCI). Methods Seventy-two healthy adult male or female Sprague-Dawley(SD) rats with body weight 250-300 g were divided into normal group(group A), acute spinal cord injury(SCI) group(group B)and acute SCI-EGb treatment group(group C). Allen 's method was used to injure segments of T8-T10 of rat thoracic spine,and Basso-Beattie-Bresnahan(BBB) scores and slope test were performed 3, 7, 21 days after operation, respectively. The in-jured spinal cords were then removed, the Nogo-A protein expression levels and changes in each phase time were detected by Western blot. Results The 21 days after operation, BBB scores and critical angle in group C were significantly higher than those in group B, and the difference was statistically significant( P〈0.05). Nogo-A protein expression of rat spinal cords was positive in each group, the Nogo-A protein at each time phases in group B and group C were significantly higher than that in group A(P〈0.05). The highest point of Nogo-A protein expression appeared in group B and group C at 7 days after operation,and decreased from the 21 days, but the expression was still higher than that of group A. The Nogo-A protein expression at each phase time in group C was significant lower than that in group B, and the difference was statistically significant( P〈0.05).Conclusion It is demonstrated that EGb can promote recovery of motor function of acute SCI rats and inhibit the expression of Nogo-A protein obviously in rats after SCI.
