摘  要:目的采用常压密闭缺氧耐受力实验联合急性减压缺氧耐受力实验,探索天麻素对模拟高原缺氧小鼠的保护作用。方法 100只雄性BalB/c小鼠随机分为两组用于两个实验,每组50只小鼠按照随机数字表法分为5个亚组:空白模型(M)组、红景天阳性对照(RC)组、天麻素低剂量(GAS-L)组、天麻素中剂量(GAS-M)组、天麻素高剂量(GAS-H)组,各10只。每日定时灌胃,连续7d。第七天给药50min后,分别比较5组小鼠在密闭缺氧环境下的存活时间以及在急性减压条件下的存活率,从动物整体水平上评价天麻素的抗缺氧活性。结果天麻素3个剂量均可有效延长常压密闭缺氧环境下小鼠的存活时间(P<0.01),降低小鼠暴露于急性减压缺氧条件中的死亡率。但天麻素低、中、高剂量组间无显著量效关系。结论天麻素能够对模拟高原缺氧小鼠起到一定保护作用。Objective To study the protective effect of gastrodin on mice under simulated high altitude hypoxia en- vironment by using normobaric hypoxia tolerance test combined with acute decompression of hypoxia tolerance test. Methods A total of 100 male BalB/c mice were randomly divided into two groups for two experiments. According to the random number table method, 50 mice in each group were divided into blank model (M) group, rhodiola crenulata (RC) group, low-dose gastrodin (GAS-L) group, medium-dose gastrodin (GAS-M) group and high-dose gastrodin (GAS^H) group (n = 10). Each mice were timed to receive gavage everyday for 7 days. The anti-hypoxic activities of gastrodin were evaluated from whole levels of mice after 50 minutes treatment at the seventh day by comparing the survival times in a closed environment and survival rates under acute decompression conditions between two groups. Results Three doses of gastrodin significantly prolonged the survival times of mice under normobarie hypoxia environment(P^0. 01), and re- duced death rate of mice under acute decompression of hypoxia environment. No dose-effect relationship was found in low-, medium- and high-dose gastrodin groups. Conclusion Gastrodin have a protective effect for mice with high altitude hypoxia damage.
