摘 要:目的探讨昆仙胶囊对博来霉素诱导的肺间质纤维化小鼠肺组织病理和血清白细胞介素(IL)-6、IL-10、IL-13、粒-巨噬细胞集落刺激因子(GM-CSF)表达的影响。方法将60只C57BL/6小鼠按随机数字法分成空白组、模型组、昆仙胶囊组及甲泼尼龙组,每组15只。空白组气管内注入0.9%氯化钠注射液0.02 ml,其余3组以气管内注入博来霉素(5 mg/kg)建立肺纤维化模型,并于造模后第7、14、28天分批处死小鼠,留取血样测定细胞因子IL-6、IL-10、IL-13、GM-CSF的含量,并取肺脏采用苏木精-伊红(HE)及Masson染色法观察肺组织病理变化。结果 (1)与空白组对比,其他各组第7、14、28天小鼠体质量下降(P<0.05);与模型组对比,昆仙胶囊和甲泼尼龙组小鼠体质量有所增加(P<0.05);(2)在第7、14、28天昆仙胶囊组肺泡炎和肺纤维化程度明显轻于模型组,小鼠肺泡炎和肺纤维化病理评分降低(P<0.05);且第28天时,昆仙胶囊组仅形成轻中度肺间质纤维化。(3)在第7、14、28天昆仙胶囊组血清IL-6、IL-10、IL-13、GM-CSF较模型组明显降低(P<0.05),而昆仙胶囊组与甲泼尼龙组对比差异无统计学意义(P>0.05)。结论昆仙胶囊能显著降低肺间质纤维化小鼠血清IL-6、IL-10、IL-13、GM-CSF表达水平,减轻肺组织肺泡炎及肺纤维化程度,从而抑制博来霉素诱导的小鼠的炎症反应和肺间质纤维化的发生发展。Objective To investigate the effect of Kunxian capsule on the pathology and the serum expression of IL-6, IL-10, IL-13 and GM-CSF in the lung tissues of mice with bleomycin-induced pulmonary interstitial fibrosis. Methods Sixty C57BL/6 mice were randomly divided into the control group, model group, Kunxian capsule group and methylprednisolone (MP) group (n=15 each). The mice in the blank control group received 0.02 ml normal saline by endotracheal injection, and the others 3 groups received bleomycin (5 mg/kg) by endotracheal injection to establish the pulmonary fibrosis model. The mice were sacrificed at 7, 14 and 28 d after the modeling, respectively. The blood samples were collected to determine the contents of cytokines IL-6, IL-10, IL-13 and GM-CSF. The lung tissues were harvested, and the histopathological changes were examined by HE staining and Masson staining. Results ①Com-pared with the blank control group, the weight of mice in the other groups decreased at 7, 14 and 28 d. Compared with the model group, the weight of mice in the Kunxian capsule group and MP group increased ( P〈0.05). ②At 7, 14 and 28d, the degrees of alveolitis and pulmonary fibrosis in the Kunxian capsule group were significantly lower than those in the model group, and the pathological scores of alveolitis and pulmonary fibrosis decreased ( P〈0.05). At 28 d, mild to moderate pulmonary interstitial fibrosis formed in the Kunxian capsule group. ③At 7, 14 and 28 d, the serum levels of IL-6, IL-10, IL-13 and GM-CSF in the Kunxian capsule group significantly decreased compared with those in the model group (P〈0.05), whereas there were no statistically significant differences between the Kunxian capsule group and the MP group (P〉0.05). Conclusion Kunxian capsule may significantly reduce the expression levels of serum IL-6, IL-10, IL-13 and GM-CSF in mice with pulmonary interstitial fibrosis, reduce the degree of alveolitis and pulmonary fibrosis in lung tissues, and further inhibit the occurrence a
