摘  要:以玉竹[Polygonatum odoratum(Mill.)Druce]根茎、茎段、叶片为外植体,研究了玉竹不定芽的诱导和增殖体系。结果表明,最适宜外植体的灭菌时间为75%的乙醇处理10 s配合0.1%的Hg Cl2处理7 min,其污染率为13.3%;最适宜的外植体为根茎,成活率为90%;经过7 d的4℃低温处理的外植体,比未经处理的分化率高出了24个百分点;在玉竹增殖培养中,最适增殖培养基为MS+6-BA 0.5 mg/L+NAA 0.8mg/L,最适宜增殖培养的蔗糖浓度为30 g/L,分化率达到了86.7%,并且植株生长状态良好,植株健壮。The rhizomes,stems and leaves of Polygonatum odoratum(Mill.) Druce were used as explants to study the adventitious bud induction and propagation. The results showed that,the optimum time for sterilization of explants was ten seconds by75% alcohol then treated seven minutes with 0.1% mercuric chloride,the pollution rate was only 13.3%. The survival rate of roots as optimum explants was 90%; after cold treatment for seven days of 4 ℃,differentiation rate of the explants was 24 percentage point higher than untreated explants. The optimum medium was MS+6-BA 0.5 mg/L+NAA 0.8 mg/L in the proliferation culture of P. odoratum(Mill.) Druce. The optimum sucrose concentration was 30 g/L,the differentiation rate reached86.7%,and the plant growth status was stronger.
