摘  要:为研究牛磺胆酸(TCA)对体外培养小鼠胸腺细胞凋亡的影响,探讨其免疫调节作用,采用梯度离心法分离获取小鼠胸腺细胞,37℃培养12 h后与不同浓度TCA共孵12 h。以流式细胞术对原代培养胸腺细胞早期凋亡进行定量检测;应用分光光度法检测了凋亡特异性蛋白酶Caspase-9、Caspase-3活性的变化。结果表明,TCA3组(终浓度为0.30μg/m L)与胸腺细胞共孵12 h后,可极显著抑制磷脂酰丝氨酸外翻,抑制细胞凋亡,TCA2组(终浓度为0.15μg/m L)可显著提高Caspase-9、Caspase-3活性,诱导细胞凋亡。说明TCA通过对体外培养小鼠胸腺细胞凋亡的调节作用来调节机体免疫功能。In this paper the study was aimed at discussing the immunoregulation of taurocholic acid(TCA) by analyzing its effects on mice thymocyte apoptosis. The mice thymocyte cells were isolated by gradient centrifugation and cultured under 37 ℃with TCA of different concentration for 12 hours. Through flow cytometry(FCM),the thymocyte apoptosis in early stage were detected quantitatively. Besides,the activity of caspase-9 and caspase-3 were detected through spectrophotometric methods. The results showed that the TCA group 3(the final concentration:0.3 μg/m L) could dramatically restrain phosphatidylserine from eversion and restrain the cells apoptosis. Meanwhile,TCA group 2(the final concentration:0.15 μg/m L) could greatly improve the activity of caspase-9 and caspase-3,and induce the cells apoptosis. It indicated that TCA adjusts organism immune function by regulating the mice thymocyte apoptosis.
