摘  要:本文采用氢氧化钾与碱性蛋白酶的新型反应体系,水解含铬革屑制备多肽水解液。以黏度、游离氨基量、氨基酸含量为实验指标,探索了酶与含铬革屑的百分比以及反应的时间、pH值等因素对反应体系革屑水解程度的影响。经一步碱水解,固液比1∶12.5,氢氧化钾用量32%,反应温度100℃,水解时间6 h。二步酶水解最佳条件为:酶用量5%,反应体系pH=10,反应4 h。In this paper,the peptide hydrolysate was prepared by the hydrolysis of chrome leather scraps in a new reaction system of potassium hydroxide and alkaline protease.The factors on the influence of the hydrolysis reaction such as the percentage of the alkaline protease to chrome leather scraps,reaction time and hydrolysis temperature were investigated by using the viscosity,the free amino content and the amino acid conversion rate as experimental indexes.The optimal hydrolysis reaction conditions are as follows: in the first alkali step,the solid-liquid ratio is 1∶12.5,the dosage of potassium hydroxide is 32%,the reaction temperature is 100℃,and the hydrolysis time is 6 h.In the second enzyme step,the dosage of alkaline protease is 5%,pH of the reaction system is 10,and the hydrolysis time is 4 h.
