摘 要:以黄瓜类质粒pC3为重要元件,构建了黄瓜叶绿体表达载体,并采用电穿孔方法对黄瓜离体叶绿体进行转化.以叶绿体总DNA为模板扩增出预期长度的外源基因,结果表明,类质粒pC3的整合没有影响表达载体结构的稳定.这种特异性黄瓜叶绿体表达载体的构建,为研究高等植物类质粒的起源、细胞器基因组间遗传物质传递等基础理论,以及推进叶绿体转化的应用提供了重要的工具.The chloroplast expression vectors were constructed using the cucumber plasmid-like p C3 as important element, and transferred into the isolated cucumber chloroplasts employing electroporation.The expected length of the foreign genes were amplified with the total chloroplast DNA as templates. The results show that the integrating of p C3 did not affect the stability for the structure of these expression vectors. Construction of the special cucumber chloroplast expression vector will provide an important tool on the research of theoretical foundation about the origin of plasmid-like, hereditary material transfer between different organelle genome and promote the application of chloroplast transformation in higher plant.
