重组IL-4R蛋白对SD大鼠过敏性哮喘干预作用的研究

摘  要:目的探索重组IL-4R蛋白对过敏性哮喘动物模型的干预作用。方法选择6周龄SD大鼠60只,采用安全随机分组法分成哮喘模型组、IL-4R组、布地奈德组和对照组,每组15只。哮喘模型组在腹部选取3处于皮下注射10%鸡卵清白蛋白(OVA)混合液1ml(OVA 100mg、氢氧化铝100mg及灭活的百日咳杆菌5×109个)进行致敏,1周后用上述同样药物再进行致敏,第2次致敏1周后将大鼠分别放入不完全封闭的箱中,用5%OVA与0.9%氯化钠混合溶液,连续10d超声雾化激发30min。IL-4R组和布地奈德组同法造模,但每次雾化激发前30min,IL-4R组腹腔注射重组IL-4R蛋白稀释液(400μg/d)。布地奈德组腹腔注射布地奈德稀释液(400μg/d),对照组用0.9%氯化钠溶液连续10d超声雾化激发30min。取4组大鼠肺组织作HE染色的光镜病理切片,观察肺组织病理变化。建立间接ELISA方法,检测实验中4组大鼠肺泡灌洗液中的IL-4和IFN-γ的含量。结果哮喘模型组大鼠肺组织有大量嗜酸性粒细胞浸润,对照组大鼠肺组织几乎无嗜酸性粒细胞浸润,IL-4R组和布地奈德组大鼠肺组织嗜酸性粒细胞浸润明显少于哮喘模型组。IL-4R组和布地奈德组肺泡灌洗液中IL-4和IFN-γ的含量明显高于哮喘模型组,差异均有统计学意义(均P<0.05)。结论研究结果提示IL-4R蛋白对过敏性哮喘有良好的干预作用。Objective To investigate the effect of recombinant IL-4R on allergic asthma in rats.Methods Sixty SD rats in 6 wk of age were randomly divided into model group,IL-4R group,budesonide group (as positive control) and control group with 15 in each.Asthma model was induced with subcutaneous injection of ovalbumin (OVA).Rats in IL-4R group,budesonide group and control group were given intraperitoneal injection of recombinant IL-4R (400μg/d),budesonide (400lμg/d) or normal saline,respectively.The pathological changes of lung tissues were observed with HE staining,the contents of IL-4 and IFN-γ in alveolar lavage fluid were detected by indirect ELISA method.Results Large amount of eosinophil infiltration in the lung tissue was observed in asthma model group,which were significantly attenuated in IL-4R budesonide groups.The contents of IL-4 and IFN-γ in alveolar lavagefluid of IL-4R and budesonide groups were significantly lower than those of asthma model group (P〈0.05).Conclusion The results suggest that recombinant IL-4R can effectively intervene allergic asthma in rats.
