益气养阴活血方对糖尿病肾病大鼠肾组织TRPC6、RhoA表达的影响

摘　　要：目的:探讨益气养阴活血方对糖尿病肾病(DN)大鼠肾组织TRPC6、RhoA表达的影响。方法:大鼠喂养1周后分为3组:空白组、模型组、中药组。建立DN大鼠模型,造模成功后空白组和模型组灌服生理盐水,中药组灌服益气养阴活血方颗粒,8周后生化检测24 h尿蛋白、血肌酐(Scr)、谷丙转氨酶(ALT)、血脂(TC、TG)、空腹血糖(FBG)等;测量肾小球直径,称取左肾重量、计算肥大指数;PAS染色观察大鼠肾脏病理;免疫组化检测肾组织TRPC6、RhoA表达。结果:益气养阴活血方能够降低实验大鼠肾组织TRPC6、RhoA的表达,减少24 h尿蛋白,升高ALB,缩小肾小球直径,改善大鼠体质量及肾脏病理,保护足细胞,降低肥大指数及血脂、血糖。结论:益气养阴活血方可能通过影响DN大鼠肾组织TRPC6、RhoA的表达,而改善上述生化指标,减轻肾脏病理损害,并对肝、肾功能未见明显损害。Objective：To explore how Yiqi Yangyin Huoxue Prescription affects the experssions of TRCP6 and RhoA in kidney tissue of mice diabetic nephropathy. Methods ： We divided the mice into three groups after feeding them for a week： blank group, model group, traditional Chinese medicine group. We irrigated the blank group and model group＇s mice diabetic nephropathy model by normal saline while the experimental group was fed with Yiqi Yangyin Huoxue Prescription. 24 - hour urinary protein quantity, Scr and ALT were measured after the mice were burdened for 8 weeks, we measured glomerular diameter and the left kidney＇s weight and then calculated the hypertrophy index. PAS staining was used to observe rat renal pathology and Immunohistochemical method was used for the detection of renal tissue TRPC6 expression. Result： Yiqi Yangyin Huoxue Prescription can reduce the expression of experimental rats renal tissue TRPC6, decrease the 24 - hour urinary protein quantity, Triglycerides, cholesterol and hypertrophy index, increase albumin, narrowing glomerular diameter, protect podocyte and improve the body weight and renal pathology of rats. Conclution： Yiqi Yangyin Huoxue Prescription may improve the biochemical indicators, reduce the renal pathological damage and there is little damage to liver function by interfering with the DN rat kidney TRPC6 and RhoA expressions.
