加减资生汤配方颗粒对肺气肿模型大鼠肺组织及血清IL-4、IL-8、TNF-α表达影响

摘　　要：目的:研究加减资生汤配方颗粒对肺气肿模型大鼠肺组织及血清IL-4、IL-8、TNF-α表达的影响。方法:采用烟熏结合气管注射脂多糖法造模,随机分为空白组、模型组、低剂量组、中剂量组、高剂量组,每天给药1次,连续给药10周。病理切片染色法观察肺气肿模型大鼠给药前后肺、支气管等组织的病理变化。采用PCR结合酶联免疫吸附法,检测大鼠肺组织、血清IL-4、IL-8、TNF-α表达水平。结果:与空白组相比,模型组大鼠肺、支气管、血清TNF-α、IL-8、IL-4表发水平显著增强(P<0.05);与模型组相比,加减资生汤配方颗粒干预组能显著降低肺气肿模型大鼠肺组织及血清TNF-α、IL-4、IL-8表达水平(P<0.05)。结论:加减资生汤干预肺气肿的机制可能与其调节肺组织及血清中TNF-α、IL-4、IL-8表达有关。Objective： To study the effect of Modified Zisheng Granule on emphysema rats＇lung tissue and IL- 4, IL - 8 and TNF -α in serum. Methods ： The rat model was made by using the smoke inhalation combined with the injection of lipopoly saccharide. The rats were randomly divided into blank group, model group, low, middle and high dose groups, once a day, 10 weeks for continuous administration. Pathological changes of lung and bronchus before and after administration of pulmonary emphysema model rats were observed by pathological section staining. The expression levels of IL -4, IL -8 and TNF -α in rats＇lung tissue and serum were detected by PCR and enzyme linked immunosorbent assay. Results ： Compared with the control group, the model group rats＇lung bronchus, serum levels of TNF -α, IL - 8 and IL - 4 increased significantly （ P 〈 0. 05 ）. Compared with the model group, Zisheng Granule can significantly reduce the emphysema model rats＇lung tissue and serum levels of TNF -α, IL - 4 and IL - 8 （ P 〈 0. 05 ）. Conclusion ： The intervention mechanism of Zisheng Granule treating emphysema may be related to regulating the expressions of TNF -α, IL - 4 and IL- 8 in lung tissue and serum.
