摘  要:目的:探讨白花蛇舌草乙醇提取物(Ethanol extract of Hedyotis Diffusae Herba,EEHDH)对人肾癌786-0细胞体外生长及体内成瘤的影响。方法:体外实验采用MTT法检测EEHDH对人肾癌786-0细胞的生长抑制作用;体内实验采用人肾癌786-0细胞实体瘤小鼠动物模型,取30只昆明小鼠,6只为正常对照组,余24只昆明种小鼠每只左后肢腋窝皮下接种0.2 m L肿瘤细胞悬液,24 h后称质量,随机分为模型对照组、EEHDH低剂量组、EEHDH中剂量组和EEHDH高剂量组,每组各6只。正常组和模型对照组灌胃等体积饮用水,EEHDH低剂量组、EEHDH中剂量组和EEHDH高剂量组分别灌胃含生药量5.0 g·kg-1、10.0 g·kg-1、20.0 g·kg-1的EEHDH溶液,每日1次,连续给药14 d,末次给药24 h后颈椎脱臼处死动物,切开肿瘤部位,剥离瘤块,用滤纸吸净其上液体,称瘤块质量,计算抑瘤率。同时分离胸腺、脾脏,精密称质量,计算胸腺指数、脾脏指数。结果:EEHDH在体外可显著抑制人肾癌786-0细胞的增值,其效果与剂量和作用时间相关。体内试验表明,与模型组比较,EEHDH低剂量组、EEHDH中剂量组、EEHDH高剂量组均能明显减轻瘤块的质量,差异具有统计学意义(P<0.05),抑瘤率分别为16.02%、31.22%和34.26%,差异有统计学意义(P<0.05)。小鼠荷瘤后,其脾脏指数、胸腺指数比较正常组显著升高,差异有统计学意义(P<0.05);与模型组比较,EEHDH低剂量组、EEHDH中剂量组、EEHDH高剂量组能显著提高脾脏指数,差异有统计学意义(P<0.05)。结论:EEHDH在体外能明显抑制人肾癌786-0细胞的增值,体内能显著抑制荷瘤小鼠肾癌786-0细胞生长,与调节免疫器官功能有关。Objective: To probe into the effect of Baihua Sheshe Cao( Herba Hedyotis Diffusae) ethanol extract( EEHDH) on human kidney cancer 786-0 cells growth in vitro and in vivo tumorigenicity. Methods: In vitro,MTT was used to detect the inhibitory effect of EEHDH on the growth of human kidney cancer 786-0. In vivo experiments using human kidney cancer 786-0 cells solid tumor animal model in mice,30 Kunming mice were selected,and among of them,6 mice were chosen into the normal control group; 0. 2m L tumor cell suspensions were inoculated subcutaneously into each left hind of the axilla of the other 24 mice. Then 24 hours later,their weight was measured and they were randomly divided into the model group,EEHDH low-dose group,EEHDH medium dose group and EEHDH high-dose group,6 rats in each group. The ones in the normal group and the model control group were perfused with the same volume of drinking water,while the ones in the EEHDH low-dose group,EEHDH medium-dose group and EEHDH high-dose group were given intragastric administration containing crude drug 5 g·kg^-1,10 g·kg^-1,20 g·kg^-1 EEHDH solution respectively,once a day,for 14 days continuously. And 24 hours after the last administration they were killed by cervical dislocation; The tumor was cut,and the tumor mass was stripped. After absorbing the liquid with the filter paper,the tumor mass was measured and the tumor inhibition rate was calculated. At the same time,the thymus and spleen were separated and weighed accurately. The thymus index and spleen index were calculated. Results: EEHDH could significantly inhibit the proliferation of human kidney cancer 786-0 cell in vitro,and the effect was related to dose and length. In vivo experiments showed that compared with the model group,EEHDH low-dose group,EEHDH medium-dose group,and EEHDH high-dose group could significantly reduce the quality of the tumor,the difference was statistically significant( P〈0. 05); The inhibition rates were 16. 02%,31. 22% and 34. 26% respectively,and the differenc
