解毒化瘀颗粒抑制急性肝衰竭大鼠肝线粒体通透性转换的实验研究

摘　　要：目的:明确解毒化瘀颗粒在急性肝衰竭动物实验中的治疗作用及是否可抑制急性肝衰竭大鼠肝线粒体通透性转换。方法:采用D氨基半乳糖联合内毒素脂多糖构建急性肝衰竭大鼠模型,解毒化瘀颗粒干预治疗,成模后24 h收集各组大鼠标本,同时观察各组大鼠48 h存活率,全自动生化仪检测血清ALT、AST含量,分光光度仪检测肝线粒体在520 nm处吸光度的变化来测定PT孔开放情况,Western Bolt检测肝线粒体与胞质细胞色素C蛋白含量,测定Caspase-3蛋白活性情况。结果:解毒化瘀颗粒可提高急性肝衰竭大鼠48 h存活率,降低血清ALT、AST含量,抑制肝线粒体PT孔开放,提高线粒体细胞色素C含量,降低细胞质细胞色素C含量,抑制下游Caspase-3蛋白活性。结论:解毒化瘀颗粒可抑制急性肝衰竭大鼠肝线粒体通透性转换,抗急性肝衰竭肝细胞凋亡。Objective：To clarify the therapeutic effect of Jiedu Huayu Granules on acute liver failure in rats and identify wether it can inhibit the hepatic mitochondrial permeability transition.Methods：A acute hepatic failure rat model was established based on D-Gal N and LPS,intervented therapy of Jiedu Huayu Granules.We collected samples from each group after 24 h and observed rats survival rates at 48 h.ALT and AST were detected by automatic biochemical analyzer and ultraviolet spectrophotometer was used to detect PT pore opening of liver mitochondrial at 520 nm.Western blot analysis was used to detect cytosolic cytochrome C protein in liver mitochondria and cytoplasm.Results：The 48 h survival rate of rats were improved by Jiedu Huayu Granules which also reduced serum ALT and AST contents,inhibited the opening of mitochondrial PT pore,increased the content of cytochrome C in the mitochondria and decreased the content of cytochrome C in the cytoplasm and inhibited caspase-3 protein activity.Conclusion：Jiedu Huayu Granules can inhibit the hepatic mitochondrial permeability transition in rats with acute liver failure,anti apoptosis of liver cells in acute liver failure.
