摘 要:目的:探讨针刺对哮喘大鼠肺组织中P-AKT蛋白表达的影响。方法:将40只SPF级雄性SD大鼠随机分为空白组、哮喘模型组、哮喘模型针刺组和哮喘模型雾化阻断剂组,每组10只。各组分别给予相应干预后,用HE染色观察大鼠肺组织形态学改变,Western Blot法和免疫组织化学法检测大鼠肺组织P-AKT蛋白的表达。结果:HE染色显示哮喘模型组大鼠气道周围有多种炎症细胞的浸润和聚集,支气管平滑肌痉挛,官腔狭窄;针刺组和雾化阻断剂组有少量炎症细胞,官腔狭窄管壁增厚程度较模型组减轻。Western Blot和免疫组化法检测示哮喘模型组P-AKT蛋白表达较空白组、针刺组、雾化阻断剂组升高(P<0.05);针刺后P-AKT蛋白表达降低(P<0.05),雾化阻断剂组与空白组无差异(P>0.05)。结论:针刺可降低大鼠肺组织中P-AKT蛋白表达以减轻炎症反应和气道重塑。Objective:To examine the mechanism of acupuncture on P-AKT expression in the lung tissues of asthmatic rats.Methods:Forty SPF male SD rats were randomly divided into 4 groups with 10 rats in each group.Groups were given accordingly interventions.HE staining,Western Blot and immunohistochemical method were employed to observe the morphologic changes and the protein expression of P-AKT in lung tissue of rats.Results:HE staining showed the airways of the asthma model group's rats had the infiltration aggregation of a variety of inflammatory cells with bronchial smooth muscle spasm and the lumen narrow but in the acupuncture group and blocking group these phenomena as above was more less significantly.Western Blot and immunohistochemical method showed the protein expression of P-AKT was higher in asthmatic model group than that of other groups(P〈0.05).But there was no statistical difference among the blocking group and the blank group(P〈0.05).Conclusion:Acupuncture can reduce the protein expression of P-AKT in lung tissue of rats to reduce inflammation and airway remodeling.
