摘 要:采用SRAP分子标记对70份万寿菊杂交后代优选株系基因组DNA进行遗传多样性分析,结果表明,9对SRAP引物共扩增得到250个位点和110个多态性位点,多态位点比例为44.0%;各引物组合Shannon多样性指数(Ⅰ)在0.054~0.466之间,平均为0.228;各引物组合的鉴别能力存在较大差异,以me4/em17、me6/em4最佳,可分别鉴定31个和42个株系。各株系获得的条带数介于168~217之间,平均为203.5条,其中多态性条带数介于22~71之间,平均为57.5条,占平均条带数的28.07%;Ⅰ指数介于0.655~0.693之间,平均为0.676。检测到株系间遗传距离变化范围为0.030~0.180,平均为0.098,变异系数介于17.81%~30.98%,平均为23.97%。亲缘关系聚类分析将所有株系在遗传相似性系数0.89处分为9类,其中株系41-3、45-1、36-2、6-1、26-2、16-1单独聚成一类,4-3、5-1(3)和35-2聚成一类,28-1、36-1和37-3聚成一类,其余58个株系聚成一类。In this study, the genetic diversity of DNA of 70 selected superior strains of hybrid progenies in Tagetes erecta was analyzed by SRAP. The results showed that 250 loci and 110 polymorphic loci were amplified by 9 pairs of SRAP primers, and the proportion of polymorphic loci was 44%. Shannon diversity index(Ⅰ) of each primer combination was between 0.054 and 0.466 with an average of 0.228. The resolving ability of each primer combination was quite different, in which me4/em17 and me6/em4 were the best. They could identify 31 and 42 strains, respectively. The number of bands obtained from each strain was between 168 and 217, with an average of203.5, and the number of polymorphic bands ranged from 22 to 71, with an average of 57.5, accounting for 28.07%of the average number of bands; index Ⅰ was between 0.655 and 0.693 with an average of 0.676. The genetic distance indexes of all strains ranged from 0.030 to 0.180 with an average GD of 0.098, and the variable coefficient was between 17.81% and 30.98% with an average of 23.97%. Clustering analysis results showed that all strains were divided into 9 groups at the genetic similarity index of 0.89, in which the strains were clustered intoa single group, strain 41-3, 5-1(3) and 35-2 clustered into a group, 28-1, 36-1 and 37-3 clustered into a group, and other58 strains were clustered together.
