疏肝健脾方药对多囊卵巢综合征大鼠性激素水平及胰岛素抵抗作用研究  

Effect of Shugan Jianpi Recipes on the Level of Sex Hormone and Insulin Resistance in Polycystic Ovary Syndrome Model Rats

摘　　要：目的:观察疏肝健脾方药对多囊卵巢综合征大鼠性激素水平及胰岛素抵抗作用。方法:随机将60只健康雌性SD大鼠分为正常组、模型组、中药低剂量组[22.05g/(kg·d)^(-1)]、中药中剂量组[44.1g/(kg·d)^(-1)]和中药高剂量组[88.2 g/(kg·d)^(-1)],采用脱氢表雄酮(DHEA)皮下注射进行PCOS模型复制。造模成功后,连续给药21 d后,分离血清,检测大鼠血清T、E2、LH、FSH、FBG,FINS水平并计算HOMA-IR水平,分离大鼠卵巢组织,RT-PCR法检测GSK3β、AKT2、GLUT4、IRS-1 m RNA水平。结果:与正常组比较,模型组大鼠血清T、LH、FSH、FBG、FINS及HOMA-IR水平显著升高(P<0.01),E2、P水平显著下降(P<0.01),卵巢组织中GSK3β、AKT2、GLUT4、IRS-1 m RNA水平显著降低(P<0.01);经过治疗后,与模型组比较,低剂量组大鼠血清E2、P显著升高(P<0.05,P<0.01),LH、FBG、FINS水平降低(P<0.05),HOMA-IR水平显著降低(P<0.01),卵巢组织中GSK3β、IRS-1m RNA水平显著升高(P<0.05);中剂量组和高剂量组大鼠血清T、LH、FSH、FBG、FINS及HOMA-IR水平均显著降低(P<0.01,P<0.05),E2、P水平显著升高(P<0.01),卵巢组织中GSK3β、AKT2、GLUT4、IRS-1 m RNA水平均显著升高(P<0.01)。结论:疏肝健脾方药能够有效改善PCOS大鼠血清激素水平及胰岛素抵抗情况,其机制可能与调控PCOS大鼠卵巢组织中GSK3β、AKT2、GLUT4、IRS-1 m RNA水平有关。Objective ：To observe the effect of Shugan Jianpi Recipes on the level of sex hormone and insulin resistance（IR）in polycystic ovary syndrome（PCOS）model rats. Methods ：60 female SD rats were randomly and equally divided into 6 groups,the normal group,the model group,the low dose traditional Chinese medicine group [22.05 g/（kg·d）^（-1）],the middle dose traditional Chinese medicine group [22.05 g/（kg·d）^（-1）] and the high dose traditional Chinese medicine group [22.05 g/（kg·d）^（-1）]. To establish the model of PCOS with Dehydroepiandrosterone（DHEA）subcutaneous injection. After the modeling and treatment for 21 days,The serum was isolated to measure the level of serum T,E2,LH,FSH,FBG and FINS,calculate insulin resistance index（HOMA-IR）,the ovarian tissue was isolated to measure the level of GSK3β,AKT2,GLUT4,IRS-1 m RNA. Results ：Compared with normal group,the serum levels of T,LH,FSH,FBG,FINS and HOMA-IR were significantly increased（P〈0.01）,the serum levels of E2 and P were significantly decreased（P〈0.01）,the m RNA levels of GSK3β,AKT2,GLUT4 and IRS-1 in ovarian tissue were significantly decreased in the model group（P〈0.01）. After treatment,compared with model group,the serum levels of E2 and P were significantly increased（P〈0.05,P〈0.01）,the serum levels of LH,FBG,FINS and HOMA-IR were significantly decreased（P〈0.05,P〈0.01）,the m RNA levels of GSK3β and IRS-1 in ovarian tissue were significantly increased（P〈0.05）,in the low dose traditional Chinese medicine group. the serum levels of T,LH,FSH,FBG,FINS and HOMA-IR were significantly decreased（P〈0.05,P〈0.01）,the serum levels of E2 and P were significantly increased（P〈0.01）,the m RNA levels of GSK3β,AKT2,GLUT4 and IRS-1 in ovarian tissue were significantly increased（P〈0.01）,in the middle and high dose traditional Chinese medicine group.
