摘 要:目的:制备姜黄素-槲皮素自微乳(CUR-QUE-SMEDDS)给药体系并建立其质量评价方法。方法:选择Capryol 90为油相,Cremophor RH40为表面活性剂,Transcutol HP为助表面活性剂,通过单因素实验优化处方,制备CUR-QUE-SMEDDS。以自微乳的外观性状、粒径分布、Zeta电位、微观形态、包封率和载药量为指标对其进行质量评价。结果:制备的CUR-QUE-SMEDDS为澄明液体,流动性、稳定性良好,遇水可自发形成O/W型微乳,平均粒径(6.90±0.09)nm;Zeta电位(-7.13±2.25)m V。HPLC法检测姜黄素、槲皮素的线性范围分别为10.728~96.552μg·mL^(-1)和1.064~9.576μg·mL^(-1),r=0.9998和r=0.9999。姜黄素和槲皮素的平均回收率分别为99.98%和100.34%;RSD分别为1.46%和1.06%。CUR-QUE-SMEDDS中姜黄素、槲皮素的包封率和载药量分别为95.97%±0.50%、(25.82±0.15)mg·g^(-1),95.51%±2.52%、(1.80±0.05)mg·g^(-1)。结论:CUR-QUE-SMEDDS制备工艺简单、包封率高,粒径分布均匀。Objective :The objective of the study was to prepare and evaluate the quality of curcumin and quercetin loaded self-microemulsifying drug delivery system(CUR-QUE-SMEDDS). Methods :Based on the curcumin self-microemulsifying drug delivery system(CUR-SMEDDS),the CUR-QUE-SMEDDS composed of Capryol 90(10.0%),Cremophor RH40(60.0%),Transcutol HP(30.0%). The optimal prescription(CUR-QUE-SMEDDS)was confirmed by single-factor design experiment,using appearance status,particle diameter,Zeta potential,transmission electron microscope micrographs,drug entrapment efficiency and the drug loadings of curcumin and quercetin as evaluation indexes to determine the quality evaluation of Cur-PIP-SMEDDS. Results :The CUR-QUE-SMEDDS prepared was a translucent and achromatous solution with good stability. After being diluted with water,CUR-QUE-SMEDDS was consisted of small spherical drops. The average particle size was(6.90±0.09)nm,Zeta potential was(-7.13±2.25)m V. The calibration curves were linear in the range of 10.728~96.552 μg·mL^(-1)(r=0.9998)for CUR and 1.064~9.576 μg·mL^(-1)(r=0.9999)for QUE. The average recovery was in the range of 99.98% and 100.34%,respectively. The EE and DL of the final prescription,CUR was(95.97%±0.50%)(25.82±0.15)mg·g^(-1),QUR was(95.51%±2.52%)(1.80±0.05)mg·g^(-1). Conclusion :The preparation technology of CUR-QUE-SMEDDS is simple and feasible. The entrapment efficiency of CURQUE-SMEDDS is good and its particle size distribution is uniform.
