摘  要:目的:分别利用丙酸睾丸酮(Testosterone Propionate,TP)联合高脂饲料及经典Poresky方法联合高脂饲料诱导大鼠多囊卵巢综合征(Polycystic Ovarian Syndrome,PCOS)胰岛素抵抗(Insulin Resistance,IR)动物模型,探讨其胰岛素抵抗(Insulin Resistance,IR)特征,并比较两种造模方法。方法:将21日龄雌性SD大鼠随机分为A组(丙酸睾丸酮联合高脂饲料组)、B组(经典Poresky方法联合高脂饲料组)及C组(空白对照组),每组各10只,A组每日皮下注射丙酸睾丸酮的注射液,连续喂养高脂饲料;B组连续喂养高脂饲料,造模第35天开始采用经典Poresky方法;C组每日皮下注射同剂量大豆油剂1次,3组造模时间均为8周。观察卵巢形态学及阴道图片改变、体重变化,测定血中促卵泡生成素(LH)、促黄体生成素(FSH)、睾酮(T)、空腹血糖(FPG)和空腹胰岛素(FIN)浓度,并计算胰岛素抵抗指数(HOMA-IR)。结果:模型A组、模型B组大鼠血清LH水平均显著高于对照组,其中模型A组显著高于模型B组;模型A组、模型B组大鼠血清FSH水平均显著高于对照组,但模型A组与模型B组之间无显著性差异。模型A组大鼠血清T水平显著高于模型B组和对照组,但模型B组和对照组之间无显著性差异。模型B组大鼠血清FINS、FBG、HOMA-IR水平均显著高于模型A组及对照C组(P<0.05),其中模型A组与对照C组无显著性差异。光镜见对照组卵巢局部形态基本正常,可见黄体、卵丘并存,两个模型组基本符合PCOS病理特征,但是模型A组囊性扩展的卵泡较多,模型B组未发育成熟的小卵泡较多。阴道图片提示对照C组有规律的动情周期,模型A组偶发排卵,模型B组动情周期紊乱,未见明显的角化细胞,提示无排卵。结论:两种造模方法均可成功诱导大鼠PCOS胰岛素抵抗模型,但经典Poresky方法联合高脂饲料优于丙酸睾丸酮联合高脂饲料。Objective :Respectively using testosterone propionate(TP)with high fat diet and the classical Poresky method with high fat diet to induce rat models of polycystic ovarian syndrome with insulin resistance(IR). And comparing compare the two kinds of modeling methods with their insulin resistance features. Methods :21 day old female SD rats were randomly divided into group A(testosterone propionate and high-fat diet group),group B group(the classical Poresky method combined with high fat diet group)and group C(control group). There are 10 rats in each group. Group A were administered testosterone daily,with continuous feeding with high fat diet;group B were fed with high fat diet while the thirty-fifth day modeling by the classical Poresky method;group C were given the same dose of soybean oil as the testosterone each day. Three group of modeling time was 8 weeks. The detection index was the changes of ovarian morphology and vaginal images,the body weight changes,the concentration of blood Folliclestimulating hormone(FSH),luteinizing hormone(LH),testosterone(T),fasting blood glucose(FPG)and fasting insulin(FIN),and insulin resistance index(HOMA IR). Results :The serum LH and FSH levels of the model group A and model group B rats were both significantly higher than the control group. In LH level,the model group A was significantly higher than that of model group B. In FSH level,there was no significantly difference between model group A and B. In T level,the model group A was significantly higher than that of model group B,but there was no significantly difference between model group B and C. In serum FINS,FBG,HOMAIR levels,model group B were significantly higher than those of model group A and control group C(P〈0.05),while the model group A and the control group had no significant difference.
