摘 要:为了研究牦牛离乳期复胃差异表达基因,试验采用健康幼年牦牛(2~3月龄)复胃组织为试验样品,以健康成年牦牛(3~5岁)复胃组织为对照,采用抑制消减杂交(SSH)技术对犊牦牛复胃组织在离乳转换期的差异表达基因进行筛选,获得差异表达cDNA文库,随机挑选96个单克隆进行测序,其中包含20个表达序列标签(ESTs)序列。结果表明∶在前胃(瘤胃、网胃、瓣胃)中发现的9个差异表达基因具有调控蛋白质合成、促进细胞生长、保护细胞等功能,在皱胃中筛选出的11个差异表达基因具有调控蛋白质合成与修饰、信号传导、激素的调节等生物功能。To study differentially expressed genes in compound stomaeh tissue of yak at weaning stage, compound stomach tissues of 2 - 3 month - oldyoung yak were involved in this experiment, with the eontrol tissues of 3 - 5 year - old adult yak. Suppression subtraetive hybridiza- tion(SSH) was employed to screen the differentially expressed genesin compound stomach tissue of young yak at weaningtransition stage ,and to obtain differentially expressed eDNA library. Ninety - six clones were randomly selected and were sequenced ,which contained 20 expressed se- quence tag(ESTs) sequences. The results showed that 9 differentially expressed genes were found in rumen,retieulum and omasum, possessing the functions of protein synthesis regulation, cell growth promotion and cell protection, etc. Eleven differentially expressed genes were sereenedin abomasum, having the functions of protein synthesis- modifieatinnregulation, signal transductionhormone regulation and so on.
