摘 要:当机体受到亲电子物质或活性氧刺激时,核转录相关因子2(nuclear factor erythroid-2 related factor 2,Nrf2)能诱导出一系列的保护性蛋白,从而抵抗内源性或外源性氧化应激造成的损害。越来越多的研究发现,某些组织比如心脏受到氧化应激时,Nrf2的活性和线粒体生物合成有关。在Surfeit 1蛋白(Surfeit locus protein 1,Surf1)突变的小鼠心脏中发现,由于细胞色素C氧化酶的缺乏导致细胞呼吸减少从而激活Nrf2。本文从Kelch样环氧氯丙烷相关蛋白1(Kelch-like ECH-associated protein-1,Keap1)-Nrf2通路与线粒体的关系以及氧化应激条件下Nrf2信号通路与线粒体之间的信号交联为重点进行了综述。Nrf2 is known to induce the expression of hundreds of cytoprotective proteins to counteract endogenously or exogenously generated oxidative stress when the body was stimulated by electrophilic reagents or reactive oxygen species. Interestingly, increasing evidence shows that Nrf2 is associated with mitochondrial biogenesis during environmental stresses in certain tissues such as the heart. Furthermore, Nrf2 activation was recently observed in the heart of surfeit locus protein 1 (Surfl) -/- mice in which cellular respiration was decreased due to cytochrome c oxidase defects. In this review,we critically examine the relationship between Nrf2 and mitochondria, and argue that the Nrf2 stress pathway intimately communicates with mitochondria to maintain cellular homeostasis dur- ing oxidative stress.
