摘 要:目的观察神经肽Y受体亚型Y_1、Y_2、Y_5在压力超负荷大鼠心肌中的动态表达变化。方法手术组采用腹主动脉部分缩窄法建立压力超负荷的动物模型,对照组为假手术组,每组6只老鼠,每组再分2、7、14 d三个时间点动态观察Wistar大鼠。无创鼠尾测血压;测全心质量/体质量的比值;苏木素伊红(HE)染色测心肌细胞横截面积;实时定量聚合酶链反应(quantitative real-time polymerase chain reaction,QTPCR)法检测Y_1、Y_2、Y_5m RNA表达;采用蛋白印迹法检测Y_1、Y_2、Y_5受体蛋白含量。结果手术组术后2、7、14 d血压进行性升高,与对照组各时间点比较,收缩压显著升高,差异有统计学意义(P<0.05);手术组全心质量/体质量的比值于术后14 d升高,心肌细胞横截面积的变化与全心质量/体质量的比值一致。手术组心肌Y_1、Y_2及Y_5 mRNA和蛋白的表达一致,与各时间点对照组比较,术后2、7 d心肌Y_1、Y_2及Y_5 mRNA和蛋白的表达上调,至14 d下降。结论压力超负荷模型建立成功,手术组术后2、7 d处于压力超负荷早期阶段,14 d进入压力超负荷心肌肥大代偿阶段;神经肽Y受体亚型Y_1、Y_2和Y_5在压力超负荷大鼠心肌中的表达呈动态过程,在早期上调,心肌肥大的代偿期下降。Objectives To investigate the dynamic changes of expression of neuropeptide Y(NPY)receptor subtypes Y1,Y2 and Y5 in myocardium of pressure overload rats. Methods Totally 36 Wistar rats were randomly divided into 6 groups,6 rats in each group:2 days,7 days and 14 days groups,each period included sham group and operation group. Operation group was treated with abdominal aortic constriction (AAC). Noninvasive blood pressure was measured by tail artery at day 2,day 7 and day 14 respectively. Ratio of heart weight(HW,mg)/body weight(BW,g) was measured. The parts of ventricles were fixed in formalin and microtome sections were stained with hematoxylin and eosin(H&amp;E). Cross-sectional area of myocardial cells was measured by using IPP6.0. Expressions of Y1,Y2 and Y5 mRNA in myocardium were detected by fluorescence quantitative real-time polymerase chain reaction (QTPCR). Protein expressions of Y1,Y2 and Y5 were detected by Western blotting. Results Blood pressure of operation group was measured 2 days,7 days and 14 days after operation,and systolic blood pressure significantly increased compared with sham group at each time point. Ratio of HW/BW increased significantly at 14 days in operation group compared with sham group. Changes of myocardial cell cross-sectional area were consistent with the ratio of HW/BW. Expressions of Y1,Y2 and Y5 mRNA and protein in the myocardium of operation group were consistent at day 2,day 7 and day 14.Expressions of Y1,Y2,Y5 mRNA and protein in myocardium were up-regulated at day 2 and day 7,but decreased at day 14 compared with sham rats at each time point. Conclusions Pressure overload model was established successfully. In operation group,there was no hypertrophy myocardium at 2 and 7 days,the heart was in the early stage in pressure overload rats;the heart had the cardiac hypertrophy characteristics at day14,that indicates the hearts had entered the cardiac hypertrophy compensatory stage in pressure overload rats.
