自拟通痹汤对实验性关节炎的作用

摘　　要：目的:研究自拟通痹汤对实验鼠膝骨关节炎软骨细胞、关节液中的一氧化碳(NO)及前列腺素2(PGE2)含量、关节软骨中基质金属蛋白酶-1(MMP-1)含量的影响,探讨自拟通痹汤治疗大鼠膝骨关节炎的作用机理,为临床上治疗膝骨关节炎提供理论依据。方法:将72只SD大鼠用关节腔注射碘乙酸钠溶液方法造模,将大鼠随机分为空白组、模型组、自拟通痹汤低、中、高剂量组及阳性对照组6组进行治疗2周,后对大鼠膝骨关节炎软骨细胞、关节液中的NO及PGE2含量、MMP-1含量进行比较。结果:自拟通痹汤组及阳性对照组较空白组、模型组的软骨细胞含量明显升高(P<0.01),关节液中的NO及PGE2含量、MMP-1含量明显降低(P<0.01);自拟通痹汤中、高剂量组与阳性对照组间比较各数据无明显差别(P>0.05)。结论:自拟通痹汤组能够促进大鼠膝关节软骨的生成,抑制关节液中的NO、PGE2的分泌,对大鼠膝骨关节炎的治疗具有积极的作用。Objective： Self-prescribed Tongbi Decoction on experimental rats osteoarthritis of the knee cartilage cells,synovial fluid of carbon monoxide（ NO） and prostaglandin E2（ PGE2） content,effect of articular cartilage matrix metalloproteinase 1（ MMP 1） content of Self-prescribed Tongbi Decoction in the treatment of rats with knee osteoarthritis effect mechanism,for the clinical treatment of osteoarthritis of the knee and provide a theoretical basis. Methods： The 72 SD rats with intra-articular injection of sodium iodoacetate solution method for modeling,were randomly assigned to the rats were randomly divided into blank group,model group,Self-prescribed Tongbi Decoction,and high dose group and positive control group 6 groups were treated for 2 weeks,on experimental rats osteoarthritis of the knee cartilage cells,synovial fluid of no and PGE2 content,MMP-1 content compared. Results： Self-prescribed Tongbi Decoction group and positive control group compared with the control group,cartilage cell content in model group increased significantly（ P〈0. 01）,in synovial fluid of NO and PGE2 content and MMP-1 levels decreased significantly（ P〈0. 01）; Self-prescribed Tongbi Decoction group and positive control group compared the data without significant difference（ P〉0. 05）. Conclusion：Self-prescribed Tongbi Decoction group can from promote the formation of knee joint cartilage,inhibit the synovial fluid in NO,PGE2 secretion,the treatment of knee osteoarthritis with positive role.
